I assume no one responded to this because no one does it? because... Spores take too long/ more work/ more chance of contams?
Is it ever useful to add some genetic diversity back into a isolate without going back to the master print/ MS

No. The whole point of isolation is to remove genetics.

Inbreeding has its ups and downs but general rule being ya want to use your earliest prints possible as they will degrade eventually

I just dunked my BRF cakes, I'm gonna prepare my bulk substrate for tomorrow. I have coir and verm ready, can awesome experienced cultivators tell me how much ratio I should use? (i have been suggested 50-50) and I don't know how much is 'one brick of coir' because my brick is kinda big and I don't really know how much of it I should use. I'm going to boil 4 liters of water and put everything together in a bucket and leave it for the night, is it cool or I should put them in pc? please, tell me what is the best way to do it, i want the best for my mushies (i love my mushies ^_^)

Id suggest about 80/20 coir to verm and around 10 qts per 66 qt tub and around 20 cakes to spawn it

I know there's not a concrete answer, but is there a ballpark timeline for how long cube LC and/or LI can be stored longterm without gas exchange? Or a definite, "The myc will die after x amount of time" kind of figure?
Like, I've always read that myc needs GE or it'll stall and die out. That's why we don't close our slant lids all the way down, etc. Is there a general idea for liquids? Will no GE kill off liquid mycelium or suspend it?

You do close off GE on slants all the way after they colonize. Theb store them.

Stare I've stored Li for 8 years and had a successful flush. But that was the strongest culture I had. On average id say 3 years. I recently revived a stored stem tissue sample sitting in a syringe in the fridge from 2014 and it fruited very well it wouldn't grow on agar, but it did grow in pf jars. I posted a thread called "how long will this culture last". Decided to put all these old revivals in that thread to see how long I can really push it. Right now I have an LC of oysters from 2007 I put to agar a few days ago. I've grown it out once a year with success. So that what 10 years old? Nature Is amazing and seems to push its own boundaries and adapt.
As for previous questions about forced albinos. Psilocybe fanaticus said he forced albanism with uv light. You can likely find the information somewhere online. But fishy mentioned he used UV light. Haven't heard back about details yet. But he will be back soon enough and if I find out ill let you know. I think the entire grow was done under uv light. I'd assume colonization of bulk sub as well.

I wouldn't make it much more than a few hours in advance. highly nutritious media that's not sterilized will mold up in a day or two

paegan said: Hey there, it is my first grow and I have questions about fruiting.
I am using a shotgun terrarium with saturated perlite and grow light 12/12, but didn't realize I needed to elevate the bin off the ground until today. Cakes have been sitting in there for almost a week now (I waited 1 week after 100% colonization before birthing) and I haven't been getting any pinning yet. It smells really "mushroomy" in there and the mycelium has overtaken much of the casing layer and still looks healthy but no shrooms...
What else can I do apart from elevate the bin and mist occasionally to induce fruiting?
Nothing just keep misting and wait!
Thanks cronicr! That is exactly what I have been doing now for the last 3 days. As a follow up:
What is a normal time to wait before I should expect pinning? I placed birthed cakes into sgfc 1.5 weeks ago (didn't mist for the first week :/ ) and I still don't have any clear signs of pins forming...as a matter of fact what exactly should I be looking for, what does the primordia look like when it is just starting out?
I have a ton of small white bumps on a few of the cakes and then I have maybe 3 or 4 much larger and totally different looking bulbous growths on a couple cakes (confused man lol)
Thanks in advance

I'm about to drill some holes for my monotube, some of the pictures have it in substrate level while others are 5" above the substrate, which one is right and why?

I did use the search function but couldn't find an answer, so thought I'd ask here.
I have a good problem of having too many colonized grains for the room and number of tubs I have. How long can I store colonized cubensis grain in the fridge and still have good success with spawning?

awesome! okay, I did my monotube and everything is done. I covered the holes with microtape for colonizing, is it okay to use it in fruiting phase too? or it has to be polyfill?

Long enough, more than months. Probably would still try to use sooner than later

awesome, some of the pictures I saw in the link looked like they are not growing as much as the ones used polyfill, is it different in your opinion? (i mean does it have pros and cons?)

Can bacteria or other contaminants land on agar and not germinate because of there only being one or two certain unfavorable nutrients, but then when expanded to grains or substrate then the contaminant shows up? Or will anything that lands on the agar start to grow?
