Of course. You will see a gamut of results. That's why we test isolates or clones and only keep the good ones. Of course ms is a crap shoot every time.

tape some quarters to your weight. it will increase pressure and decrease water loss. just keep an eye on your temps when you do.

its def possible. part of the huge amount of work that is finding a monoculture entails doing many many test grows to find the right genetics.

it is very possible. Don't think I have ever came across that, but it is definitely possible.

Hey!

I am a complete beginner, and just recently started reading seriously about mushrooms. I have my hands on some dried golden teacher.. What would be the best way to try and start a culture from this? I do not know how they were dried.

Depending on how they were dried you may be able to reanimate them with antibiotic agar. However if they were dried with high heat it would be pointless and even if not it would be a challenge. Personally it would be far easier to just buy a syringe or print.

Yeah problem about that is not too easy to get in to my country..
Is there any way to tell from how they look, how they were dried?
What kind of antibiotic agar is suggested?

You can get gentamiacin to make antibiotic agar or buy tetracycline from the pet store. But you cannot pressure cook tetracycline so it needs to be carefully added during the cool down before you pour your plates. It's a bit tricky.

I don't recall hearing about your country banning spores. Should be easy to order some.

They banned it in 2013.

Anyways thanks very much for the replies.

Think this should be enough to get me googling for the night..
I do not have a pressure cooker at hand, is it essential.
Or is it possible to do this with just steam ?
If it is essential I will let it wait until I get one.

ok say i have ~50 dollars and no PC,but i have cubensis spores on syringe and want to grow them and dry them for later (obviously). how would you guys go about that?

Is shaking your grains still necessary with cakes or anything with a casing? I have verm layers but I imagine shaking would ruin that.

Shaking should pretty much only be done with grains. At the very least it needs to be done in an airtight container with a nice filter. If you have a dry verm layer as your filter, or have an open substrate with a casing, shaking is detrimental.

I've always seen spore syringes made the following way: scrape spores into a shot glass, spray sterile water into it, and then aspirate/spray 3-5x before capping the syringe.

Assuming there's a reasonably clean print, including the foil casing, is there a much bigger risk for contamination if you simply dropped the foil into a jar of sterilized water for SHIP aspiration?

Obviously spores are inherently dirty and there's always risk in creating syringes from them, but with a properly cared for print, any reason not to try this? It just seems easier overall and can make more syringes. The only other issue I can think of is maybe the spores won't dislodge without scraping initially.

Follow up question: I doubt people making syringes for vendors are going 1 shot glass at a time. What's the common method for bulk syringe making?
