A lot of contaminations might not grow on agar and will on something else. Or vice versa may do well on agar and poorly on grain.
Total plate count agar used in QA will grow a lot of aerobic organisms.
For what its worth PDA and MEA and YPD all grow a lot of things well.
Things that spoil beer are anaerobic and very specilaised to tolerate the high abv and hop bitter acids. Theres very special media incubated anaerobically to grow these microorganisms. But not all media grows all spoilage bugs. So many breweries use 2 or 3 different beer agar recipes to plate finished beer on so that hopefully if there is any bugs it will grow on one of the recipes.
Theres a big brewery in town that had an infection they were completely blind to since the two agar mediums they used to test against didn't grow the bug that was in their beer. I got a yeast sample from them and it tested bad. Low and behold it only grew on BMB agar which they didn't use. They ended up having a million dollar product recall because the spoiler eventually over carbonated bottles. It would have cost them so much less if they stopped the problem earlier than spending money on cardboard boxes, six pack holders, bottles, crowns, labels, labor, refrigeration, shipping, and then having to reship them back, and destroy them.
Rather than dumping it out before even filtering it.

So would grain water agar be best for the purpose of finding these things.
Since it's the grain were growing on anyways wouldn't anything that would grow on grain be the same for the agar? And vice versa.

Likely. Thats the idea behind diy beer agar. Theres other media they make. Such as universal beer agar UBA it has things like tomato extract to help encourage colonies of beer spoliling lactobacillus to form since they like to grow in beer but not on agar a lot of times

Nope just make PCR cheaper and cheaper. Agar for QA will be old news in a few years once even mom and pop places can afford something that gives results in the same day vs waiting 7 for incubation
Agar will still have use but no one will be trying to find spoilers with it in industry
Even universal beer agar isn't universal they're always trying to make a better recipe that grows every spoiler known

That's just not true. tuberculosis wont grow on any of our agars, but theoretically could get on one. Same goes with anaerobic bacteria, if not all of them die they still may be transferred around on aerobic plates. Which is a problem growing yeast on ypd then using it for beer in which a unseen anaerobic organism can later find itself in an appropriate home

That’s mainly why I was just curious, because I blew heavily on one plate and spit on another a few months back just to see what would happen and surprisingly the one I blew on didn’t grow anything. The other one looked fucking gross after 4-5 days lol

Streptococcus would not grow well on our plates if at all. But you could spit on a plate and it could get there. Theyre usually facultative anaerobes so they'll likely form no colony. And would thus hide on typical media
Hospital labs work would be a lot cheaper and easier if they only had to use MEA

Wait, what? I remember RR's video advocating storage after Parafilm with the a bit loose, but Frank snaps his air-tight. I remember researching more and seeing RR and others saying cutting off all air will kill the mycelium. Found another post with a guy comparing slants of Parafilm and air-tight and air-tight ones didn't regrow. Is this horseshit?

I've never closed off my slants to GE. they are just wrapped in parafilm and put in an insulated lunchbox in my fridge. seems to have worked fine for my slants that I keep for ~a year or so.

Hey Guys,
I went out and bought the biggest tub i could find, althou i believe it might be too big...
Size : 79.5cm x 58cm x 43.5cm / 2.6ft x 1.9ft x 1.4ft
I would like to use this as a fruiting chamber for BRF PFTek Jar cakes & Mini Flat cakes. Do you think it will work out ? Let me know your thoughts.
2nd:
-Mini Flat cakes: created by mixing new subtrate with fully colonized mycellium jar grated up in a tray.
^^^ Is this a good method to do? I am limited to one syringe, and this seems worth it if i can double the cake size with only a extra week of waiting.

Hi. I made a mono 45l box. P.cube Ecuador ratio 1:2 spawn - coir. no date stamp (bravo) But the most it could be standing at 28C is 14days. I did not trim the liner, just inverted the edge on all sides of the box and covered with lid for GA.
How does it look? Thats water droplets covering mycelium.

Question about grain spawn inoculation. My first time inoculating jars (WBS), it was with a TEK that used a Tyvek filter on the outside top, and 1 single small hole with a self healing silicone port. So basically, after you pull out the syringe, the jar is basically sealed with no GE. The jars actually colonized nicely, but I don't understand why, since there was no method of GE. Without knowing, I did a G2G transfer using this same type of jar/filter, and the mycelium grew a little bit and then completely stalled. After waiting another week, I did some reading, and decided to drill another hole in the jar and then cover with micropore tape.. There was a gush of (air?Co2?) that either came in or out of the jar, once the lid was punctured. Then the jars proceeded to colonize.
So I was just curious why the initial jars were even able to colonize with a spore syringe in the first place. It seems like they shouldn't have been able to do so with no GE. Thanks.

Should I knock up my agar straight from the syringe or use my makeshift inoculation loop I made from a bread tie? I've never used one bc I'm kind of a newb and don't know how.
