Right. That's how I keep my slants.
bod, you've slanted species air-tight for longterm storage and pulled them out with no issues down the line? Most other TC's and common knowledge of mycelium seems to say they require oxygen or they suffocate so this is confusing.

I've done slants for cubes twice ever then decided why bother. I've done a bunch for yeast because I had to. which was always air tight after colonizing for a few days.
as I learned the idea is that since just above freezing in a fridge you're trying to halt metabolism or slow it to the fractions of what it would be otherwise. so the little air in there is enough for a year theoretically. Then you're supposed to transfer slants under normal refrigeration ever 6-12mo. I find petri dishes last that long or longer anyway.
petri dishes in fridge lasts long enough for my needs.

yeah ive always found pasty plates in a ziploc bag in the fridge can keep for very long. just pour em a little deeper.
same with cube slants. i dont keep many. my slants are for a tamp clone and a bunch of edibles. the only cube one I have is muda's original Envy-SS culture.

Im getting back into the swing of things and wanna play around with a few substrates and boxes, g2g and pc .5 gallon jars and I'm having a hard time finding info on pc jars that big. Any info would be great Thanks J

2-3 hours at 15 psi will be fine. make sure you have enough water to run it that long - check your manual.

Heyo guys.
So I have a thought. If you had a plate you believed had a hidden contam could you add sterile water to it and transfer that to a new plate to see if anything grows.
If it was contamed with a bacteria you can't see well. I feel that the bacterial colonies would be taken by the liquid. Then on the new plate grow out before any Chuck's of mycelium do.
Thoughts?

a good way to isolate cube myc from bacteria is the agar sandwich tek. no link needed, just make some agar plates, take a normal transfer, than open up a clean agar plate and either drop it on top of the other or cut out a piece thats big enough to cover the wedge you transferred. like the name implies, you want to the sandwich the transferred wedge between two pieces of agar.
this works because the cube myc grows filamentous strands through the agar, and can grow up and through it to the top. bacteria, however, cannot. it will only spread on the 2d surface and cannot penetrate the agar. so by growing through the agar, the cube myc can escape the bacteria.

Thanks to you and bod for the additional info regarding slants, liquids, and fungi breathing and shit. I could post this in true confessions, but I'm very bad at differentiating between very tomentose mycelium (esp starting from MS) because I got decent enough at agar early on and kept the same cultures alive for years (still have like 12 unopened syringes that are probably dead now).

That's Ghostie from a really nice Shroomery member. What do you think, Munch? Bacteria for sure?

thats a tough one. I've had some plates do the thing where the myc dips below the surface of the agar. but your surface level myc looks a bit different. you said those are spore inoculation sites? or are they transfers?
 this was kss that i eventually cleaned up and fruited just fine

The "clear" growth on the leading edge is a good sign that its healthy imo. It may not be pretty but that looks like mycelium to me stare

Hi, I am from Mumbai, India. I wish to try magic mushroom in micro doses under supervision of a medical doctor. I tried getting it here in Mummbai (Bombay) but unfortunately was'nt able to get it. I even tried mother tincture of psilocybe cubensis. I am suffering from panic disorder and I wish to use it as a medicine. Could someone please enlighten?

Yeah, those were from spores. My loop has a very small threaded head that dips into my agar below the surface sometimes. I'm thinking that's what has promoted the unique growth pattern. Definitely going to take a transfer to a new plate and see what develops.
