Does this look like trich to anyone else? I tried germinating a PE swab of mine by tearing off a two fibers and burying them in the plate. This growth popped up 3 days after inoculation in between the two spots where I put swab fibers. I think I know the answer :facepalm:

Also, please forgive how nasty the plate lid looks. These plates got horrid condensation and I'm a moron so I decided to turn them upside down and shake the plates side to side to clear the lid of condensation. I know better, but like I said - I'm a moron.

probably cobweb / Cladobotryum mycophilum, or could be a whole host of white molds.

could be alright though, take a transfer and see if it gets better. Sometimes germinations are really weird looking I guess from all the genetics fighting it out. But that doesn't look crowded it just looks off.

Rw x twc germinating, finally some clean plates to work with.

I'm also wondering if anyone has had issues with bacteria using yeast in agar?
All of my yeast plates had bacteria everywhere but plates without yeast are spotless.

Thanks for that, tormato :smile: I've got a tendency to be pretty hard on myself. But yes, I'll take a couple transfers and let y'all know how it turns out! I just got worried because I'm not really sure how to be sure if it's mold or mycelium. Thanks for the help!

It's growing. Honestly I find it's morphology changes depending on the foods. Who gives a shit if it's clean it's clean.

Grain water is all I use now.

You'll learn through experience how to identify and distinguish between the two. Looking back at your original post I would wait a little longer before making a transfer...3 days seems fast, but not impossible..time is a good tool when identifying molds and myc... typically molds grow pretty fast compared to mush myc. Post another picture in a couple of days and let's see what ya got! :cheers:

Mother in law from hell over — only had time for basic maintenance. These plates grew out! I think one of the pins is pinning?

Pouring plates in the morning. Would you transfer anything specific or just a whole lot of different transfers? Or just go to grain?

I don't know the cause but this doesn't look great. It's dying or something as it grows and has the floccular morphology. Maybe viral. I wouldn't play with those cultures any longer and seek new spores from a new source.

Those two originated from pins taken from jars pinning in vitro and were washed in a cap of bleach:pint of water mix.

I’m already planning on taking spore prints and some more fresh tissue samples so will stick to those!

Ah I see.

I don't wipe clone material with anything, especially from a jar or plate. The mushroom is literally a dense sponge of mycelium fibers, any liquid contact will soak into it. You can't really wipe anything off it for mechanical removal and any sanitizer will be soaking into it, and that can't be good.

I would just lay a plate pin whole, or rip the mushroom in half and take internal stem/cap tissue for a full size mushroom. I have extremely good success with cloning and never wipe.

Good to know! I was paranoid about the pin somehow being dirty coming from a not-so-perfect BRF jar -- hence the bleach. I think I got that from watching somebody pick a mushroom while I was hiking when they pulled out a little jar with some bleach water and popped a stipe into it.

I've had *awful awful terrible* luck with agar plates thus far (probably 30-40 plates in with no success* but I'm nowhere NEAR giving up hope. A lot of attempts at MSS -> Agar that just proved so ugly and bacterial. I have a really wonderful variety of MDK that has been a profound trip and I don't want to lose the genetics. Although it might be small, the next few fruits that come off the cakes are slated for prints and tissue samples to plates. I'll do several as it's my first time going tissue -> plate.

From what I've read and from watching videos, it seems like good sterile technique in a SAB, ripping the stipe like a piece of string cheese, and a red hot (and then cooled in agar) scalpel to take a little tiny piece seems to be the procedure? I really would love to get a clean culture from these MDKs (and for practice making clean plates from all of the other varieties I have going!).

yep you've got the idea right. If you squeeze the base of the stem to separate the fibers a bit and tear towards the cap you can peel the whole stem and cap in half easily. Then you can make 2 cuts of the stringy stem tissue and lift some fibers out easily. Just do your best to keep your fingers away from the area your going to sample. SAB is great for taking tissue samples because there is no airflow blowing the possibly dirty stuff off the mushroom.
