Step 1:



Gather Your Materials. 1 gallon jug of distilled water, a scale, funnel, foil and light malt extract powder. Also 5X 1L media bottles and magnetic stir bars with stirrer and 25qt AA PC will be used.

Step 2:



From the jug of water, pour out about 300ml into one of your media bottles to make an air bubble.

Step 3:



Weight out 18g of LME powder.

Step 4:



Pour the powder into the jug using a funnel. Make sure you poured enough water out in step 2 so the funnel stays out of the water.

Step 5:



Put the lid on the jug and shake the hell out of it for a few minutes. Try to get all of the powder to dissolve. Let it sit for a half hour.

Step 6:



Turn the jug upside down and shake it to dissolve anything that settled. Work it all out. After you get all the sediment mixed up add your 300ml back to the jug and shake a little more.

Step 7:



Use the funnel to pour about 800ml into each media bottle.

Step 8:



Add your stir bars and put the lid on.

Step 9:



Tighten lid and back off a quarter turn. Cover lids with foil.

Step 10:

Step 11:





Place in your pressure cooker, vent for 15 minutes and sterilize at 15 psi for 15 minutes. Allow to cool naturally, never drop the valve or weight to release pressure. I cool in front of my flowhood for a few hours before removing the lid of cooker, tightening the lids and placing on a rack to finish cooling in the hood.

Step 12:



Inoculate the jars with a small wedge of colonized agar medium. Before removing from the hood, loosen lid about half a turn and wrap the neck with parafilm. This keeps the neck sterile for pouring the LC into your bags or jars.

Make sure to tilt the jar to get the wedge in the water if it stuck to the side during inoculation.

Wait 24 hours for growth and begin stirring vigorously twice daily for 30 seconds or 24/7 depending on your setup.

Yes works great. I've heard of people use as little as 0.1%.

Nice, I will have to try that myself to compare :thumbup:  Just getting back into it after a 6 year break, I had to go back and search what people use.  Most I ran into said like 4% but that seemed high to me.  This search always comes up with stuff that's way too old...

Ive tried LME between .25% and 2% and havent noticed much of a difference except for the color of the LC lol. Nice writeup sandman!

Usually when I run LC its only a 200ml solution and I have my lids tight during colonization. Do you leave the lids loose because its a larger volume and they can potentially stall without the GE?

Yea anything over 2% and it gets crazy dark, even 2% is really dark.

I leave the lids loose but wrapped, I find they grow faster and results in a larger mass this way.

This points towards gas exchange. Begs the question: what’s the right amount for LC. If loose > tight, modified ought to be better than unmodified. Yet the various teks are divided on that topic.

I'm trying some out with tyvek and a large hole, I think like 5/8", we'll see how that works out.

How much colonized do you want before using? Can I apply LI logic and have small amount of mycelium to water just shaked up?

Can I do small amounts like 100-200ml and let it colonize a few days then shake the shit out of it and use? Or is it a high risk pouring uncolonized nutritious  liquid??

It should be well colonized before using IME. It takes about a week or less.

Gotcha, got a half pint with .5% lme going to noc up this weekend. I have 6qt pc rn so no media bottle for me atm…

They have some on the zone. They are fairly cheap and bulk but they are cloudy. It's called a reagent bottle.

Sandman, I’m totally new to liquid culture but this seems like this tek I could handle. As far as inoculating my grain goes. How much LC is needed per quart jar and what is your inoculation procedure. Just pouring in front of a hood? Thanks for your help.

Yes pouring right into the spawn bag or jar is how I do it. Just try to get as little of a slosh in there as you can, should be about 10-20ml for a jar.
