Ps. cyan - Made 3 xfers which will go to grain.

Mazatapec T2 - She's lookin' ropey. I'm not quite sure if I should do another xfer or not.

Your cyan culture looks great. When did you transfer to that particular plate out of curiosity?

Thanks. This is my favourite plate so far. I believe it's T3 from print.

It was transferred 30th of Sep so about 11 days old at time of this photo.

The T2 looked amazing but I wanted to expand the culture.

The new transfers will be going to grain.

In my experience, Maz is very ropey... also very, very side pin-y... heads up!

Id take a few xfers from those thick ropes from 10-noon... see if you get more consistent growth.

What do you guys think is better?

A few sterile water baths vs h2o2 for a dirty plate pin?

Been holding onto this one RWxMM plate for months. Parafilm cracked and I tried to clean it up but failed. Don’t know why I kept it but it’s just pinned a nice chonker. Totally white fruit but fat like PE.

If the pin is big can't you just split the stem in half and take a tiny chunk out from the middle of the stem?

I've dunked in iso and h2o2 with success. Multiple samples on multiple plates would be smart.

Check this out. It's a very round (melmak) pin growing upside down.I think it might have originated  from a knot on the side of the petri dish then began growing downwards.

Also some nice Mel×PEU germination plates

A few of obvious sectors to go after on the right plate.

Nice. Did the stone originate from a jar or a plate?

I've been holding onto these for a bit. I wanted to make sure it was viable before posting it.

Blackstrap Molasses.

Blackstrap molasses has about half the sugar of LME and approximately 1000% of the various micronutrients, namely calcium, iron, copper, potassium, magnesium, manganese, selenium and vitamin B6. Many of these minerals are dissolved as salts, making them more absorbable than say, in the form of rock gypsum, dolomite, or even the cellulose filled hulls of grain.

Blackstrap molasses agar
8g blackstrap molasses
10g agar
500ml Brita filter water

The plate pin has mold on the plate - sterile technique problems on my part. However I like the sectoring happening on the one plate.

The gourmets love this stuff. Chicken of the woods and ganoderma Tsugae.

I also did a couple of LC using 1.5g BSM to 500ml filtered water. I would do 1g or .5g next time. It's obviously dark in color, but the clarity is better than any LME broth or grainwater I have ever made. No sediment whatsoever, and I think it's because the BSM starts as a liquid?

PF redspore on the left. It's in a jar with a filter I normally wouldn't use for LC, so I am afraid to swirl it too hard. APE on the right, broken up nicely.

I would love to see someone experiment with this stuff who has a hood and real plates - the condiment cups are ok but you can't get the full picture.

My LME broth is crystal clear at .1%, I've found that lower nutrient levels result in better overall performance.

The lower the nutrient content and the lower the dissolved solids in the water the more clarity your broth will have. I use distilled.

Higher nutrient levels trigger mycelium to slow apical hyphal extension to exploit the abundance of resources, lower available resources triggers an exploratory phase with increased apical extension. This is one of the reasons why low nute agar is used to outrun contamination.

That's why I like the BSM, much lower nutrition than LME at about half with all the weird stuff to support healthy growth, no need to add peptone and yeast and this and that.

I wish my .1% LME came out like that. I use maltoferm but I get solids in there.
