That explains it! I still have yet to make it past that weird chalky looking MS mycelium with APE :/ Do you usually place your transfer pieces mycelium-side-up, or down? Does it matter?

I've got some nice Yeti and Ghost plates growing out now I'll share in a few days just to keep this thread on track as a picture dump thread :lol:

Hey guys I'm having constant issues with bacterial looking mycelium in agar plates. Every time I go to rye the jars turn just slightly bacterial. The entire jar colonizes, but with little yellow/orange dots of bacteria in a couple spots on most jars.

My test jars I leave sitting out are fine showing no signs of contam for weeks.

I'm guessing it's got to be my agar. I'm not sure why, but all of my transfers show this weak, non rhizo growth. The myc changes and "raises up" after I do a transfer. I've gone way past t10 with these strains and they always look the same. All white, but bacterial when put in jars. You can literally see the grain directly near the agar wedge/myc turning slightly bacterial while the rest of the jar looks fine.

I'm thinking it could be my agar. When I PC for 45min, using just LMEA and Agar/Blue dye, I still see little dots of what looks like LMEA clumping in the agar. Even when I tried boiling the water first like d3monics tek.

Anyway, I'm trying to make some antibacterial (antibiotic) agar, or purchase some if possible. I plan to try these plates along w/ some premade agar and see if I see a difference. These plates definitely show weird growth, and are always bacterial no matter how many xfers I do.

Can anyone link my to a good Antibiotic Agar tek? Only one I've found seems pretty old.

Pictures are worth 1,000 words. It sounds like your sterile technique is to blame to me :shrug:

Contaminations can be sneaky little bastards so it would be best if you could show us a picture of your plates before you use them

A few of those look fine, you’re probably caramelizing your agar a little at 45min. I’d suspect it’s when you’re transferring your agar to the jar. What’s your process?

I sterilize my rye grain for 2 hours to be safe (used to do 90min but I'm trying to figure out this bacteria problem).

Agar plates are 500ml h20, 10gLMEA and 10g Agar, 2 drops Blue food dye. Even though I sterilize 45min I still see little clumps in agar though? I feel like people have said they only get this when they use yeast.

I sanitize my flowhood area/gloves and usually take about 1/2 of a fully grown out agar plate and cut it into about 6-7 pieces, xfer them into the jar, shake the jar up, then flame my scalpel and repeat.

I've stuck my scalpel into test plates and no contam. Maybe I should put some regular, uncolonized agar into a jar? Idk what other tests I could run that I haven't already besides that. Test plates and jars left out seem to be ok. 

My grain always passes the paper towel test before loading into jars. I'm so confused honestly.

I am wiping down jars thoroughly with alcohol. I normally do wipe down the plates but sometimes I admit I do forget that. Could that be causing the odd looking growth whenever I take a transfer? I notice yellow bacteria dots in the jars, but never on the plates. Looking at those plates I posted above, would you say that looks like weak mycelium fighting bacteria/contam? You can notice more irl the mycelium in the newer plates looks weak when it first grows out, like really weak. Then it seems to start to consolidate in this really tomentose form, and when I take an xfer, it changes and seems to raise up a bit which I'm sure is a bad sign.

Do you take care to not pass your hand / wrist / arm over the open plates?

Do you set the lid out of the flow stream? The sweet spot, I'm told, often does't go all the way to the edge or to the bottom. I have less contam now that I have a half-inch high wire cookie plate to do everything on. And I spray that with ISO before starting.

How do you seal your plates?

Have you tried without the blue die? Some red food dye can slow shroom mycelium growth, which maybe could keep it from out-competing bacteria? Who knows.

When was the last time you changed filters? front can last a long time, I replace back every 3 months.

I pour plates, seal them up, and let them sit a week before using to see if they have any contams. I eliminate about 1 contam plate every 100 that way. Was more earlier on.

You can pour a lot, let them sit a week, seal them, then put in the fridge for months until you need them.
