Active Pluteus Miraculix potency results * 3
    #27877805 - 07/26/22 11:28 AM (1 year, 5 months ago)

Welcome one and all for another installment of "How strong is that mushroom?"

On today episode we will be testing the Pluteus genus, specifically Pluteus americanus or possibly Pluteus saupei,  species is hard. Quantitative testing is done using a Miraculix Q test. If anyone is unfamiliar these tests they are easy and quick to use, somewhat inexpensive, and requires only 150mg of dried material either concentrate or fruit bodies. The accuracy of the test is +-10%.

Click here to bring you to the original find and identification.

The samples were very fresh, only being dried and collected one to two days prior. They dried quickly at 155f for 8 hours until cracker dry.

150mg was weighed out consisting of both stipes and caps to homogenize the sample.

The sample was ground into a fine powder

Ground sample mixed with extraction solvent for 10 minutes with intermittent mixing

1.0ml of extract drawn and filtered

1.0ml of extract is added to the analytical reagent which is placed in a hot water bath (80-95c) for 15 minutes.

After 15 minutes a color change reaction has occurred indicating the presence of psilocybin.

The reagent color is compared to the included color chart indicating a potency of ~0.9% - 1.0% or 9-10mg psilocybin per gram of dried material:mushroom2:

Side notes-

The smell of these mushrooms both fresh and dried smell almost identical to Psilocybe cubensis. They have that psilocybe sweet farinaceous smell whereas the common non-active Pluteus have a offputting, dirt type odor.

The bruising was evident within a couple minutes of damage if not faster.

Most Pluteus I find are growing on decaying logs or larger sticks but these were growing from well rotted hardwood duff or dust on the forest floor.

Samples were found late July 2022

I have Panaeolus cinctulus and Gymnopilus luteofolius lined up as the next to be tested. If anyone has a suggestion for other species they'd like tested then please leave a comment letting me know.

All testing is done out of pocket for the sake of science and curiosity everywhere. Thanks for reading!

This is the citizen science we need!!!! Good work and appreciate you sharing! 1% is pretty stout for non-psilocybe too!

Mid-late summer seems to be the season for these. The only P. americanus I found was early August last year in Maine (which should be added to the Maine list!) was quite small and hard to spot, but it had that distinct sweet smell while other Pluteus just smell like soil. Excellent work on these, wish this species wasn't so hard to find and grew in bigger clusters!

Does anyone know how reliable this test is? Because this came up recently in another thread where someone got a false positive result on a non-active mushroom using the Ehrlich test, but that test will give a positive result when there are other alkaloids in the mushroom, such as tryptophan, not just psilocybin.

So far it seems to be fairly accurate and targets only psilocybin. I believe Alan said he compared it to LCMS and it was within a small percentage accurate to the Miraculix. I could be remembering this all wrong or just talking out of my ass.... But I swear Alan or another mycologist agrees it's accurate.

I have heard that the test can return false positives from other tryptamines, like serotonin or 5-HTP. Not sure how reliable that information is. However it is likely that many mushrooms, both active and inactive, synthesize non-psychoactive tryptamines as well.

Regarding accuracy, HPLC results agree with the Miraculix results to within 10%.

No one knows if there are mushrooms that give false positives - it would be interesting to test a whole lot of inactives and see if any test positive.

A few people have tested a few inactives (oysters / button mushrooms) and it didn't detect psilocybin, but broad screening would be interesting.

All Panaeolus species are said to have tryptamines like 5-htp and serotonin, however my friend tested a bunch with thin layer chromatography and only found non-psilocybin tryptamines in certain members of the Panaeolus papilionaceus group.  It's not known known if these tryptamines would test positive with a Miraculix test.

This was a thought of mine as well. Although somewhat cost prohibitive. Specifically common Pluteus cervinus to test against. It could also be applied to Panaeolus as Alan mentioned.

Another question I've been thinking of bringing up is do the tests only test for psilocybin? In their literature it only mentions psilocybin. I'd like to believe it's only testing for one but if we're questioning how well it can differentiate between indole tryptamines it makes sense to ask.

Thanks everyone!

Specifically the part concerning how results can be influenced by many factors. I'm sure this was included in part as legal protection.

I'd personally love to have a list of known cross reacting substances if Miraculix has one. I would assume during testing they must have run across that type of problem.

Other than the price I wouldn't mind testing known inactive species looking for interfering substances. If enough people are interested I'd consider doing it. Maybe set up a citizen science testing fund? Ha for now relax and enjoy until the next installment of

Did you ever eat any of these pluteus? Trip reports are few and far between. Wish I would have seen this thread months ago, i got hit with long-covid so I only went foraging a few times, collected a few pluteus and gyms.

Thanks! I've been using Nigella sativa oil, agmatine, NAC, ibuprofen, acetaminophen. I tried loratadine, made my inflammed sinuses worse i think. What worked for you? I got long-covid at the very beginning, my whole family had a mysterious illness in January 2020. Was getting better then got messed up from the pfizer jab, pretty much identical symptoms. Covid again last Christmas... was finally starting to do a bit better then I got Omicron this past July... been having all sorts of cognitive issues again, anxiety/depression, severe joint pain, nerve pain, racing heart, fatigue... I'm starting to get strength back in my right hand, it swelled up really bad and was in severe pain. I'm supposed to have surgery, maybe I don't need it right away tho. I'm finally getting some sleep and feeling a bit better, in less pain. It's a weird disease that's for sure.

God damn dude you got hit too many times! Sorry to hear that. I had covid once this year, back in the spring. Shitty flu-like symptoms lasted about a week, really achey and sick-feeling. But the weird effects lasted months after, like the loss of taste/appetite, and most strangely, a blurred-vision and photosentitity thing which also lasted months. Made me really think about how diseases can affect us!

Around the same time a colleague of mine had 2 different versions of the flu at the same time, and he was worse off than I was, lol. There's some crazy shit out there and bad luck can strike, unfortunately.

It sounds like the anxiety that it's caused you hasn't made things any better either-- have you considered first seeking out mental health treatment?

Yeah I've been talking with other long-haulers on reddit, there are thousands on there, seems to be a good resource for info, much of reddit is a cesspool though lol I've been learning about the micro-clot theory, histamime response, immune diseases, how inflammation affects the brain etc.
I've been doing some neurotherapy stuff to help me function better. I think originally some of my symptoms I thought were just from psych meds so I got off a bunch, also got off suboxone and phenibut, almost off gabapentin. That probably hasn't helped my anxiety. I think part of it too is anxiety from my heart racing and brainfog.
Luckily I still managed to collect and grow enough mushrooms the past year that I'm still good until next season. Maybe I'll try some pluteus or gymnopilus tea soon. They might be more potent than I previously thought. Some of these pluteus I've collected bruised pretty heavily.

I really wonder what is in the Miraculix tests that provide such reliable results. From what I've seen there's just a corrosive sticker on the package. Does it have a particular smell?

At first I thought it was some kind of Ehrlich reagent variation with an iron salt added. But now I'm thinking more in the direction of cinnamaldehyde/HCl, or perhaps nitrosonaphtol (1-nitroso-2-naphtol ?). Both were known reagents for psilocybin in the old days of paper and thin layer chromatography.

Maybe I ought to order some and have a look at it during the coming winter months.
