test tubes agar

does anyone know if you can sterilize agar inside a test tube or will it spill over? and if not how the hell do you pour into them?

that did not answer a single one of my questions and i could have dione that search myself, matter of fact i did, though didnt bother to read the threads because they didnt seem to pertain. i read them now because i figured that you may have actually known what was in them and they may say something about preparing slants, however after reading them all and not really learning anytihng new, i now see my first assumption was correct, THEY DO NOT PERTAIN TO THE QUESTION. thanks anyways. i still need to know if slants can be prepared in apressure cooker with agar already in them. the reason i asked was because i didnt see an answer on the boards so i am assuming it isnt there, i could be wrong, if someone knows of a link that might tell me i would be happy to read it but please dont send me something that DOESNT PERTAIN and expect me to read it. thanks, no offense

Use google if you cannot find your answer - here.web page
You can PC a slant tube, with agar in it. Just don't release pressure & allow it to cool over night - in the PC. If you release pressure FAST - While its still VERY HOT, agar can boil over.

thank you so much. i didnt mean to be a dick or anytihng. i had googled and what not i just wasnt having any luck. that is exactly what i was looking for. " You can PC a slant tube, with agar in it. " this is what is going to determine whether i buy a bunch of disposable petri or a set of tubes. thanks for helping:)

Half pint Canning Jars & (Fool Proof) Potato Agar
? Grate 1 potato into 2 cups distilled water.
? Boil for 30 minutes, strain, add distilled water to make 1 Liter. ? Add 2 tbsp agar, 1 tbsp dextrose or corn syrup, 1/2 tsp nutritional yeast.
? Simmer while stirring slowly for 10 minutes. ? Pour into 1/2 pints (about ? to 5/8 in. deep)
? Place on lids, not tight, and pressure cook at 15 psi for 30 minutes (cooking too long may/will caramelize the sugars).
? Cool gradually so you don't boil the agar. Tilt pressure cooker as it cools so you will get a slant on the agar. (if you want one)
? Once cooled, tighten lids & tape over lid hole with alc swabbed Scotch brand clear storage tape.
? When ready to innoc (in glove box or flow/hood), remove lid, innoc & replace lid.
? Remove storage tape & replace with polyfil, or millipore surgical tape, or tape TYVEK over lid hole.
In a few days, you should see growth

Funny I just got PMed on this one. Quote:

heh, yeah thanks hyphae that was me. i really was talking about isolating into tubes and then expanding culture amount from there.
