Totally get it, Stipe. I don’t wear gloves because I haven’t noticed any different in the SAB in contam rates for agar work, which are functionally zero. If I worked in a lab I’d be wearing gloves too lol. But for my application, washed hands and proper angles accomplish my goals well within parameters I think everyone here would find acceptable.

If you’re getting lots of contaminated plates, I’d examine your entire sterile procedure, not just where a gloved hand is making contact with the underside of the lid, which doesn’t come into contact with the media at all. Not that that isn’t the problem, exactly, but if your contam rates are that high I’d start with how you PC it and work forwards.

like plate is 90% colonize and at the edge green mold appear its still white didnt turn green yet?

start over? or cut it out?

Yeah, I wouldn't fuck around with the risk of inhaling mold unless absolutely necessary. If I see mold it's an automatic toss.

If it's still just a tiny spec, make a transfer, but not if it's sporulating or occupying a large surface area on the plate. Like the satellite mold on the previous page, that's fine for opening, imo.

what it looks like to me, mold spore landed on plate. and its a tiny spec of white but i have seen green mold so many times i know in coming hours it will go green. i think it came from oranges going bad running air purifier now

I had this problem with glass plates, until I moved to disposable sterile plastic plates. Glass plates I had didn't quite fit together at all points around the circle as well as plastic ones too.

Is there any reason why you switched to disposable from reusable glass? Why not reusable plastic cups?
I specifically went with a glass, because it is autoclavable, but the lack of a seal/presence of a gap is a dealbreaker for me to buy any more

i never had had issue with glass plates using for three years and counting, i did found some batch of agar having strain of e.coli that does not die and it was track back to vendors of aliexpress.

in coming hours this bacteria might appear on agar too.




thing about glass petri-dish is you need to tightly pack it with parafilm or pvc film, its meant to have a gap, that gap is used when you pour agar to let the excess moisture gases escape, otherwise it in autoclave the glass petri-dish accidentally wet and there is no way for steam to escape it will explode.

Do you have a specific way of storing your blank/unused plates before inoculating them? I parafilm mine once being used, but i assume that gap is what allowed contams to get in them during the few days I left the blank ones on my shelf in my grow tent.
I’m confident it wasn’t the agar because the contamination would of been on all of them but the ones I sealed with parafilm immediately were fine.

once you pour agar and it settles
dish are ready to use, if you don't cover them with paraflim contam's will take over.
you have to make sure all of them use and unused are cover with paraflim. otherwise contam from air will enter and colonize.

i keep mine unused store in airtight bucket like this all of them are with pvc film(join the pvc film gang)
all are glass and cover with pvc film

You fellas must use way more plates than I do. I wrap individually because 20 plates will last me like 2 months.

Wow I wish I only used 20 plates a month. When I was doing a lot of work I was going through almost 150 per month. Just to get a bunch of cultures out to the community I went through 140 in 3 weeks plus what I normally used wish was almost a total of 200. It sucked cleaning that many plates but I love the holy grail plates. I wouldn't be able to afford this hobby if I had to buy disposable petri dishes.
