Tried out the Maltoferm 1001, had already tried the CBW golden light and from a chart I had found the difference between the two was that the maltoferm is food grade while the cbw is brewers grade.

Having trouble finding that chart but by any means, did 2% nutes 1.375% agar, following the table mycolorado posted. Definitely the clearest unstrained agar I've had to date, very stoked to get rid of the boogers :biggrin:

Wondering if the grade is responsible for the hot break somehow as the brewers grade is supposed to be higher quality, so its not a quality thing in a bad way lol

I am wearing gloves I've sprayed with ISO alcohol (I think everyone does).

I re-spray them a few times during a session if I'm doing a lot. I spray into the room, not onto my work area. (though I spray and wipe my work area before I start).

I spray both sides of one glove, set the alcohol down and rub my hands together to get alcohol all over both gloves.

I hold my hands up (not over the work) and dry them with the flowhood after I re-spray, and never use the flame torch while I'm wet or even just after they've dried. (If you're using an SAB, never use flame inside it).

I pick the lid up with my fingers down (like a claw machine), then gently flip my hand over so the lid is upside down, and kind of toss it onto my work surface.

I make sure I don't pass my hand over the inside of it or especially over the open plate.

To put it back on, I reach under it, palm up, grasp it, flip my hand over again like the claw and place it on the dish. Then use grating tape to seal it, a few times around, then one time around with micropore tape, gently squeeze the tape shut around it to close any gaps.

Doesn't sealing it involve picking up the whole thing and not just the lid?

You can try the celltreat gripring petris - there is a slightly pronouned ring on the bottom plate that the lid rests on.

You can also ditch the gloves and no need to constantly soak everything in alcohol. You can wipe down your surface with it, run the hood for a few min before working and wash your hands well and you’re golden.

With or without gloves I'm going to be getting my fingers under the lip of the cover to pick up the bottom. No good. But I like your "slide it to the edge of the table" idea.

And here's a photo of one of my plates. You can see where I streaked it (looks like a "Z I"), but something is growing in between the streaks, so would I be correct in assuming that that is contamination?



I'm still trying to figure out how to take clear photos. I tried a white paper background but the contrast was actually worse.

No need to tear the agar when you streak….gently slide your loop across the surface.  If you didn’t streak in that spot then yes, it’s likely a contam.  The technique I described is fool proof.

I don’t glove up and don’t worry too much about where my fingers touch the lid, since the lip isn’t going to touch the agar. It might be an air quality problem?

I keep my hands well away from open vessels, but plenty of other's don't. My tools are long, which prevents hovering, but I use a hood which creates turbulence around tools and vessels which could carry particles into your work.

I hate to always be that guy but I'm a stickler for proper tools and techniques. Lab techs wear gloves for a reason, you'll never see one of them raw dogging their work.
