Just one last thing: Anyone that can give me any input about the sterilization time using a 10 PSI PC? I've seen a chart with the times, but for the grains. I'm gonna search for it, but any info would be appreciated.

A liquid media is much easier to sterilize than grain.

I do 15 min at 15 PSI. I know someone who uses a standard pot to sterilize and does 60 min. I would assume that 30-45 min would be enough at 10 PSI.

For grain you def want to refer to that chart.

Thanks for the input, Rusty. Considering the data on the chart (the usual time people use on 15 PSI * 2,67), 40 minutes should be enough if I take your 15 minutes as reference of the "right" time on 15 PSI. So 15 * 2,67 = 40,05.

I'll search a bit more on the matter, but I think this is a good starting point.

But I never said it was the "right" time. I said that I do 15. I know some here do as little as 10 min and others as much as 45 min at 15 PSI.

I don't get contam at 15 minutes so I just figured why do more. Also note that I boil small media bottles of 250ml content per bottle!

I'm sure you'll be fine at around 40 min

Yeah, my first search was for PP5 plastic containers to use but here where I live I just didn't find anything of which I'd sure that wouldn't melt / ruin inside a PC. All the brands and items you find around here are from the USA; for expample, I couldn't find any Ball jars or any wide mouth ones of the right capacity. : )

If you're going to progress beyond pf tek, I'd recommend instead of tape on 4 holes, drill a 3/8 inch hole and use synthetic filters. Link in the article linked in my sig line "things I wish I'd known."

Also if you're PCing agar, let the PC shoot steam 10 min before putting the stopcock on. is noisy as hell, I wear headphones. Then put the stopcock on using tongs so you don't burn your hands. Then let it get to between 15 and 20 PSI (is easy to keep at 18 PSI on mine), turn the heat down to like 1/4 and it should stay at that heat. If not, ride it until it does. You'll get it after a few times.

THEN start counting your time....20 to 25 min for agar or LC, 2 hours for grain is what I do.

Then let it cool until pressure is zero before opening.

Don't put a jar in with the lid screwed tight, may implode, or suck in contams as it cools.

I use a jar with lid screwed tight, but has synthetic filter, which also works to equalize the pressure.

I had a bunch of invitro pins in one of my pf jars and I did my best to remove them without disturbing anything and ended up transferring a little verm that was stuck to them and a couple has 2-3 little pins.

But the growth from them has been awesome to watch. I put these on these plates 2 days ago and each one has these nice white spikes of myc shooting off in every direction. These are Lipa Yai pins from this jar:



Here they are 2 days later plated up:









Give em some time and they'll hopefully grow into some decent cultures to take a transfers from. The other ones got eaten, lol. These little shits actually had enough kick for me to feel it too. Hopefully it's a good sign of their potency.

Going to go to grain with the transfers within the month and have a tub made up and fruits between Halloween and Thanksgiving. My cakes should be fruiting in the next week.

I used a lower nutrient and softer agar recipe this time. I ordered some yeast to start adding to my LME agar. Hopefully will help speed growth too.

Finally got my ghost clone to stop throwing insane sectors. It was wild. Around T3 one of the plates was split evenly down the middle heavy rhizo and silky tomentose. Happy with how it’s going now.

Sometimes when I separate the tommy stuff away from the rhizo stuff the tommy stuff gets a lot more robust on it's own. The colonies have loving relationships and express loving dominant and submissive roles. The tommy one liked the rhizo one so it took the back seat on purpose.

They wall (even when sectoring) when they don't like each other.

Hi folks!

A few weeks ago i boarded the agar ship and i would appreciate if you could have a look at some of my plates.
I started with two spore prints. #1 is a expensive GT print which shows a more or less weak growth from MS to T2. I expected more for the money.
#2 is a print from a MS-grow of mine. I label it "???", because i'm not completly sure what it was, but very likely EQ. I took this print just for fun and under not so sterile circumstances, but this little fucker shows pretty strong growth.

The plates to the right look ok to me. The left ones have a questionable sector, i guess you know what i mean. The sector on the lower left one appears somehow yellow, but this must be a light issue or the table shines through or whatever. In reality it isn't yellow, but looks like the one on the upper plate.



Is this mold or just thin myc?
It is not unorganized, grows as fast or slow as the mycelium, but i'm absolutely not sure about it. Maybe one of you has an idea.

I like this one.

These are the plates of the GT-print. i don't really like them as they are so slow and weak growing.
Would be cool if you could share your thoughts.

I did this for nearly a year with various jar contams until I:
1. Started doing the 10 min steam shoot
2. Replaced micropore tape with synthetic filters.

I tried microfil, but it didn't work for me. Some people make it work, are able to have it work into a hard little pill, but for me it got poofy again and was problematic.

Some people make micropore tape work for bulk tek too, but not me.

Look into filters. About as cheap as microfil, and perfect. They work for many runs through the PC, and just never have to think of them again.

I sand a little where I put them before I place them, and wipe the dust from sanding off. I don't sand much, like 5 or 6 scrapes on the top of the jar with light sand paper. I also put high-heat silicon around them, but I've heard that isn't needed.

But that may contribute to them being able to go through PC more times. I've run them all through probably 20 times and they're still holding well.

Hey y'all! Just wanted some opinions on my first agar attempt... Be gentle, I'm a noob.

These are attempts at getting an isolated batch of myc from dried fruit (I know, not steril and risky). However i was able to get some decent growth without any contamination from my first inoculation to the agar. I then took pieces off the good looking myc and transfered to new plates. I'm now having random growth and im not sure why. All these transfers came from the same myc from the same good inoculation. I have two that look good and a few that look wild. Can someone please elaborate on what i am seeing here? And possible reasons for the randomness?

1st two pic look like mold to me. Last 3 look like myc growth. Maybe a seasoned agar adept or TC can chime in.
