Where should I take my transfer from? This will be my first attempt at transfer, this is a clone from my APE cake grow. I’m loving the way it grew out on agar

So y'all ain't putting 500ml in your 500ml media bottles?

I put 500ml + volume of dry ingredients. :justdontknow:

I prefer 250ml bottles but I started buying 500ml since they are close to the same price and hold more volume.
I guess I could just start putting 250ml in them. I don't get boil over in the PC though I run a lot of water in PC when doing agar and it doesn't seem to be a problem.

If something is working for you, there is no reason to try something that sets you back just because another person says they prefer it that way. Enjoy your 500ml prep and only change to improve!

I do 400 ml in my 500 ml bottle just for the headroom.

I is a clone on it's second transfer....II is from a spore print no transfers I'm new to this does the clone look fine?

Does grain water agar need to be pressure cooked longer. Recently poured a bunch of plates and got bacteria in maybe half of them. I've used grain water agar in the past but I used to PC for 45 minutes this last run I PCed for 15 minutes.

I know it's probably just bad sterile tek especially since I am way out of practice. Just hard to believe I lost so many plates.

Also is it necessary to cover arm holes while cooling in a SAB? I didn't used to but maybe I should start?

My plates are also getting a little on the old side. Packaging is still in great shape, but maybe they are out of date. I think they have been in storage for 2 or 3 years now.

In the past I would loose maybe a single plate every 10 sleeves. This run I lost around 50 plates after pouring 100. That's not a passing grade... By a long shot.

15 min Def not long enough. I run mine @18psi for 30min.

You have to vent, that's a standard.
Agar only needs 15 minutes in the pc.
