Greetings all. Haven't starting growing yet and starting off with AGAR.

Read somewhere here that it might be a good idea to use a wheat AGAR recipe (will be using wheat as grain) so it gets used to wheat as a nutrient source.

Any truth to this, and if so a wheat recipe I can start off with?

Ordering 4 cube syringes next week.

A few weeks ago i boarded the agar ship and i would appreciate if you could have a look at some of my plates.
I started with two spore prints. #1 is a expensive GT print which shows a more or less weak growth from MS to T2. I expected more for the money.
#2 is a print from a MS-grow of mine. I label it "???", because i'm not completly sure what it was, but very likely EQ. I took this print just for fun and under not so sterile circumstances, but this little fucker shows pretty strong growth.

The plates to the right look ok to me. The left ones have a questionable sector, i guess you know what i mean. The sector on the lower left one appears somehow yellow, but this must be a light issue or the table shines through or whatever. In reality it isn't yellow, but looks like the one on the upper plate.



Is this mold or just thin myc?
It is not unorganized, grows as fast or slow as the mycelium, but i'm absolutely not sure about it. Maybe one of you has an idea.

I like this one.

These are the plates of the GT-print. i don't really like them as they are so slow and weak growing.
Would be cool if you could share your thoughts.

Greetings to the community.
This is my first post and first ever try to grow ....
Yesterday I made a transfer from spore swabs to agar, I have four plates and here are the photos of the best and worst ones (imho). The first one is a glass petri dish dry sterilized in an oven, and the second one is a catchup container rubbed with isopropyl. So it seems to me I have some healthy mycelium in the first picture (although I have some worries about the white edges of the agar, I dont remember that moved swab there). The second one seems totally spoiled with something like a cobweb or something else.. almost all the surface is covered with a light white substance, and the region near the swab is clear... Am I right? Do I need to worry about white edges in the first plate and make transfer for example.. or it's better to wait now to see the dynamics...?

You should not wipe things with iso to sterilize, iso does not sterilize it sanitizes.

If you're making pp cups then you must either  go for no pour or sterilize the pp cups separate and then pour.

I’m about 10 months into this hobby, not a veteran, but a couple tips I wish I had started with:

You don’t need more than 1 cube syringe to get started. But having more won’t hurt. But definitely do get at least 1 Natalensis syringe. You’ll thank me for this.

Also LME agar seems to be the standard around here. And I definitely prefer it.

Quick question: anyone ever get like nutrient boogers show up on their plates beneath the agar? I made some MEA a few days back and there’s little booger colored strands of something in there. Sort of hair like but small. Could it be malt extract that didn’t fully dissolve? I did put a clump that hardened from ambient humidity into the bottle.

I poured 20 plates a few days ago. WBS water. 2 of them have a small black dot at the bottom, below the agar.

I just assumed it was some residue in the grain water as it was there from the pour and didn't grow.

I am not sure if something similar can happen with ME :confused:

Yeah it looks a bit like that but they’re pretty small.

My worry is that it’s bacteria, but it’s clearly on the bottom of all plates and besides that it’s only been a couple of days. Doesn’t look like it’s growing either. I should take a pic when I get home; it was silly to ask without pictures lol.

But yes it looks similar.

So I'm not sure what you have going on with your dish but it can tell you what's up with mine. It was the last plate I poured. The agar was starting to set and I had to tilt the plate so the agar would cover the whole bottom of the plate. It left that "hair booger" mark.

I hope this help and is also your case and don't have contam.

Here’s a couple of examples of what I’m hoping are ME boogers. A day or two after pouring seems way too fast for random bacterial contam under the agar.

I get those in my ME too. Right from the PC I can see them.  Not sure if it is carmalization causing something to fall out of solution, or dust that fell in the ME. 

I have a LME LC that looks like that (I am doing a bulk LC test with CM, BRF, and LME LC jars.  Probably overkill making 1.5L of LC)



I am optimistic that some of that might be mycelium, but I think the dark spots are pre PC contaminants.  I think a way to get rid of them might be dissolve the LME and filter before adding agar.
