The HBO Mak swab had a 66% success rate! Granted, 1 is moving significantly slower than the other but 2/3 germ rate is pretty darn good for a shit grower like me I feel like!

The KSSS bacterial pin clone sample ended up outperforming the bacteria and quite literally swallowed it and I’ve now got plenty of usable material for a clean transfer! So stoked I’ll be able to save a failed grow considering I pumped 10 cc’s of spores into an AIO bag in all of my infinite noob wisdom. At least I don’t have to buy more to try again.

Found a few neat cultures from my AMVPxTBC swabs :dancer:  So far all the test tubs have had both normal fruit initially and then fin clusters start showing up about the time you're harvesting the first flush of normal fruits.

Hey everybody, just wanted to post a dish and ask if this is plate pins happening and then I see the post above.

I guess it's clear it's plins, but what do I do with this.

I left the plate to throw it out since it looked sketchy. Now this.
Can I give it some water or is that a nono?
If yes how?
What do I do with the plins? Do I put it to grain or can I experiment with these in some other way?

I think these are just specks on the outside of the plastic, but it's just about the best time to transfer anyways, so I will go ahead with that.

Other than that, do you feel somewhat confident that this is indeed cube and not just random mold contam? :smile:

I am considering using the low nutrient agar for taking transfers from a germ plate or from a plate with some contamination. I know I should take my transfer far from Amy obvious contamination just seeking an expert opinion on low nutrient agar. I've been battling some contaminated jars and want to get the cleanest mycelium possible to eliminate it as a source of contamination. I will also be PCing my jars for a full 2 hours from now on.

The 1-inch white circle in the middle? If you noced this plate with cubes, that is cubes.

I can't guarantee 100% there are zero contams in there, since some can be invisible with naked eye. But it looks right and I'd transfer it.

If this plate didn't have those possibly bacterial dots that you think are plate imperfections, of if you are 100% sure they are on the outside of the plastic, not inside, I wouldn't rule out sending a doughnut of the outside half-inch of the white circle to grain.

But if those dots are inside, I'd do one more transfer. And, you know, even if the dots are on the outside, if you want your plates to look like a SmellyHobbit plate.

I just poured a few agar cups and I evidently cut some corners that made my cups a little different. I used barley malt extract and there’s a lot of particles in it. How would I filter those on the next batch?  Would a coffee filter work after it’s been heated and mixed?

I put some spores to the cups I made and I see a lot of growth (not contams, out of 25 I had 4 that had mold and one with bacteria, I kept the bacteria since I’ll transfer that soon but the mold ones I just tossed). Do you all typically try to get every growth separated to make sure there’s no contams we can’t see or do you just send it to grain if there’s nothing showing just to see which ones fruit?  I sent some mss plates to grains but they’re cubes and the next batch I have other species so I was thinking of sectoring until I get clean growth. Only thing with that is I made like 3-4 cups with each strain and it’s looking like 10+ colonies on each so I could end up with a LOT OF CUPS lol.

I’m also not sure if the growth in seeing is growth from one spore or if they’ve mated, am I being ridiculous thinking that the growth I see might be monokaryotic?

Thanks man. I’ve been working on a different project for a few months. I just cloned these last week and the plates I got were so interesting I think I’m going to put the older project on pause and see how these do.

Those are more than 50% full, contam or weird shit would have showed up by now.

Also fuck me, I did like 10 T3 transfers and they all seem to be less organized than the mother plates. :justfuck:

Many people would let that grow out half again as big, but most people don't let it grow all the way to the edge to transfer.

Point of transferring at this point is to get clean mycelium, and to get good looking, well growing mycelium. You can do both at that point you show there, but some people wait. I don't see the need to. You've got organized rhizo-y looking mycelium on the edge, and it looks clean. Waitging longer lets any hidden contam grow out.

I let it grow out more on the T2 or whatever I'm sending to grain, because I want more mycelium to send to grain. I usually do 7 quart jars at once, because that's what fits in my PC.

But if you're looking for transfers, I don't see the reason to wait.

I would do it now, but nothing will be harmed by letting it grow 2 or 3 more days (depending how fast it's growing.) 

Anyone who replies contradicting all this, I believe is showing
a personal preference, not science, and should "show their work" and say why they let it grow out more before a transfer.

Growing all the way to the edge can lead to contam too, if you're plate isn't entirely sealed.
