Have you folks not yet noticed that every recipe works? One recipe will not work more or less with regards to culturing fungi, common bacteria, molds, or yeast.

Very true, right now I'm simply trying to diagnose why my cultures aren't looking right.
The plates work, the mycelia grows, but it looks tepid and weak, this makes it hard to pick out from other molds or contaminates.
The grain I put them on produced fruits - but the agar itself produces tepid weak growth. 
Only reason why I'm focused on my recipe.

I didn't have you in mind, I was specifically referring to the above reply. Grain water has been the only exception ime, it "works" but produced thin growth whenever I've played with it.

All other common agar recipes at 2% nutrient/gelling strength serves as very reliable media for anything we're likely to culture.

Keep in mind that most other additives like peptones, yeast, etc, are meant to sustain organisms for the entirety of their life cycle while on agar. Those organisms aren't transferred to other media types or substrates like we do with our cultures.

Gotcha, Thanks so much for your advice really, it has been a huge help with figuring out what's been up with my plates.
Most likely I believe the old Malt Extract(it's almost over five years old) and the use of Grain Water are my true problems here.

I was having terrible growth on old LME and purchased some briess LME on the zon and continued to see terrible weak growth. I doubled the nutes to 4% and it solved the issue.

Typically, the standard recipe of agar and nutes at 2% should yield good results whether you’re using dog food, LME, brf, etc.  I’m not sure why this brand of LME was yielding poor results but bumping up the amount made a serious difference. I think what stipe is saying is there’s no need for overly complex media concoctions to get good growth.

I know, I just couldn't resist.  I've seen recipes with 6-⁷ ingredients and I agree, that is unnecessarily complicated and not needed.  I have a bottle of peptone, so I might as well use it if it won't adversely effect my agar.

Sounds like a plan I'll do up a larger nute batch as well just to have it ready for testing.

For some added detail this was noob β€œblack” agar I made with food coloring rather than activated charcoal.  Learned why no one does it that way….





You will notice the same thing when putting more than 1 transfer per plate. They kind of stay away from one another but in reality they are so close together they are touching the growth even if it is not apparent on the surface. If you leave them long enough they will go all white with no sectoring.

I noticed that even with the same transfer piece, if plop it off-center on the petri and then move it around to get it nice and centered, there's a good chance I'll get zoning effects.

What gives :psycrankey:??

I've had great success with agar in the past but now I'm wondering if my damn city water is fucked up or something of the likes. I forgot recipes called for distilled water because I've been using tap just fine for years but I will be pouring more plates with strictly distilled water from now on (just a disclaimer)
But still, check this out and let me know your thoughts because damn

I started these 4 water agar plates 5.19, ~7.5g agar - 500ml water


19 days out this seems like extraordinary little growth, previously these cultures have performed well

I started these plates on 5.22, 2 different cultures put to WA(blue), MEACA(purple,silty), and a Honey agar plate (maroon, made off-the-cuff with no nute measurement, might have mollases in it, was super high took 0 notes)





Ironic to me that the agar I wing and don't care about is the one that shows any signs of growth

Looks like good germination.  I’d pull from a few different sections and work with them all. You’ll find many different phenotypes that way hopfully.

I've been cleaning house today, culling the petri horde and I'm wondering what to do with a bunch of germination plates and some mediocre T1's.

I'm thinking of putting most of the germination plates in the fridge and leave some out alongside the T1s to see if I get any plins.

What do you guys do with your old germ plates and so-so T1s?
