Altford, maybe I misunderstood your intentions. If you are not trying to isolate a strain I think you are already good to go for grains.

Is the satellite, contam or tamp myc. Place your bets. Answer revealed tomorrow.

Looks like myc placed by the drag of your scalpel blade during transfer. If true the answer would be a satellite colony but not contaminant.

Back on my bullshit once again, with new varieties to try out.

Cambodian: These dishes went to grain on the 16th. Started from a print from my lockdown grow. I still have a few prints that I've thought about giving out, but burnout + life hit me real hard before I could test and send. Other than being on the wispy side when it started (which tweaking my agar nutes for the T2s seemed to help with) it seems to be doing pretty well nearly three years down the road.

Ricky:Someone crossed TW4 and PE6 and somehow wound up with a RW-like specimen. Seems slightly slower than the Cambos, but still growing well. Going to grain as soon as I get more jars and prep (would've been yesterday, but the shipment was delayed )

Air Force 1: This one is interesting. There seems to be very little info on here (only two other threads mention it, last I searched) and the place I got the syringe from said many growers had difficulty getting these to fruit. Two germination plates took just under a week to show visible growth, and it's super tomentose, to the point I'm wondering if it's not the myc I want. T2s are still really fluffy, but the growth rate and organization seems to match up with cubes... I dunno.

APE: First PE variety, and it's taking its time like I heard to expect. This is the only germ plate out of four to have visible growth to this day, + two of its three transfers.

Silly question, is it worth taking a transfer from a plate with a small trich colony growing on it or is it a waste of time.

Yup. That’s kinda the purpose of agar, yeah?

I was concerned they once the trick sporilates the whole place is fucked.  Ok, I'll give it a go and see what comes of it.

Thanks Mr Hobbit

Ooo sporulation. I feel the ick factor from here.

I’ve done it. I feel like I remember some people saying if they see that verdigris color they don’t even open the thing, but I’m not that paranoid. I’ve transferred healthy myc, AND I’ve messed around with removing the contam. Had success with both, but I think cutting the contam out is probably not a wise move.

Yeah, the more I thought about it, the more I didn't like it. I didn't want to open the cup anywhere near the rest of my stuff. I decided to cut my losses and toss them. Made new transfers from other clean plates. Better safe than sorry.

Totally get it. Easier to just make some more healthy tissue than fight the mean green should it get out of control 👍

Cross from Hobbits Challenge.

I made transfers from these today. They're pretty dry, I'll make the pucks thicker and more hydrated when I do them for the challenge.

If you want something that is uniform every time you will need to continue transferring from each of those plates, with the goal being to make a transfer until there are no more sections in the mycelium. When you have an individual strain of mycelium, it will grow uniformly.

If you're saying what I think you're saying, that's not how any of this works. If you're saying what I think you're saying, it's a common misunderstanding.

Where did you hear this info?

If you changed the word "sections" to "contaminations" and ditched the last sentence, the part of yours that I quoted would make sense. (not saying those plates had contam, I didn't look at them, I just got confused by what you seem to be saying.)

Can you post a pic? Curious to look at it. Also if you have any pics from before it sporulated, that'd be great.

Sorry man missed your reply. Some of them are starting to look ok, some of them are utter trash though.

Lol, thanks!

The plates were only 4 or 5 days old so no great loss. made new transfers from the original transfer plates today after I scrubbed the shit out of my SAB and everything thing that went into it including my hands and arms.

I think you are the one mistaken. Strain isolation is not that complicated. Once you have an individual strain, it grows uniform and without sections.

Or do you not know what a strain is?

You can transfer a dish a hundred times and it will still keep sectoring and going from rhizo to tomentose and back again. You can't isolate anything other than clean mycelium when doing agar.
