What do you cut it onto? The 1st plate (agar would look like WW2)? The inside of a plate cover?

more often than not I'll just jam the whole damn swab into the agar lol. Send a T1, if not germ plate wedge to grain

But that only works so often lol

Can someone tell me what is the white grow around the swab on the yeti plate?

For me it looks kind of like bacteria. But I would wait to hear what more experienced members have to say.

Is there any chance that the mycelium will outgrow it so he can make a transfer?

I don't see mycelium, only bacteria. I would say it's a lost cause... :frown:

Yes, give the plate time before discarding it.
He has a fair bit of real estate on the plate for growth so it can grow and he can transfer some clean mycelium.

But it has been two weeks since he inoculated it. I am not so sure that there will be any mycelium growth. In my opinion the bacteria has already taken over the swab and keeps the mycelium from growin.

(But that is just a assumption, I could be wrong - so don't listen to much on what I say, I don't want to give false advice. It's how I would handle this case)

I've had plates germinate after a few weeks that overtook bacteria giving me ample starting material to transfer.

Also, it doesn't cost anything other than a tiny bit of space to just leave the plate and check it once a week.

It's bacteria but again that plate was posted to show.. bacteria.

I don't need to bother cleaning it because I already have another one that is clean.

Here - from the exact same qtip swab:

Edit: @Shrips - if I really had to, I bet I could let it grow and manage to clean it. I've succeeded with worst. Here have a look :

Yeah, got them from a really awesome member here <3  And I shared a bit of the print with an other member to spread the love

I am really happy how clean that print was, I can't see any contams from the inoculation.

:whathesaid: Yeah, that dude is pretty awesome. We must have both started them at about the same time. Your growth is faster than mine, but I'm still trying to get my GWA on point so I'm not surprised. I'll be transferring to a new batch soon, so I hope it picks up some speed then.

GERMINATION Y'ALL!!!

Sorry for the excitement, my kids don't give a shit about microbiology yet, and I don't share this hobby with anyone else in my life, so you all have the privilege of dealing with my excitement here.

A couple lessons learned, I think.

First, I'm pretty sure my agar is too hard, that's why the first five plates I tried anything with did nothing. Did 10g agar, 10mg LME, 500ml and pre-boiled before PCing (learned on this thread that last step was a poor idea).

Second, pre-stretching the parafilm seems to be SOP. The parafilm has split on a handful of plates, so will be trying the pre-stretch next time.

Will also be trying 7g agar instead of 10g to 500ml water, see if softer agar works out better for germination.

Otherwise, here is a pic of an APE multispore plate I knocked up this past weekend. Of the three plates I hit with these spores, this is the only one that is showing signs of life. Also did three with psilocybe cyanescens, and three with pan cambo, and none of those have shown any signs of doing anything.

Just wanted to celebrate here a little and say thank you to all you fine people that have posted so much information, lessons learned, etc for decades. It's much appreciated! I'm working on a setup for better plate pics as well, so promise I'll quit dropping these bad ones here soon
