P.E.

This was a swab plate that was really dirty but the myc came back and ate everything. Plate was inoculated late September.

You guys know how soot is sterile and you're not supposed to wipe tools down after flame sterilizing like some folks over on the YouTube do.

this is the 2nd time ive tried doing somegerm plates lately with No germination :frown:

This time , i even took ppls suggestions and made the agar very soft and wet. 8 grams agar per 500ml water.

Can someone give me some tips how to make these ma fuckas germinate? lol

I hear ppl say that it needs extra heat to trigger , but i hate how condensated it gets when put into a warm area.

In the past when ive done spore plates, theres always been germination with in 1-2 weeks.

THe spores im using are no more than 2 years old to.

Any helpful tips would be highly appreciated !

What can sometimes help the germination is that you make a "small" MSS and keep it for a couple of days at roomtemp (no need for fridge) do help the spores with the germination. Especially when the Sporeprint is so old.

Edit: you said you only put agar in there and no LME/ME? My Agarrecipe is for 500ml Water, 8gr Agar and 8gr LME.

Maybe also try Rottens Let's talk about Water agar, this may help aswell.

MSS = multi spore syringe?

Im reading some stuff now on making syringes.

oops, its MEA not pure agar. when ever ive used a spore syringe the germination is quick, maybe its nessecary for hydration.

Someone once told me to make an agar mix with activated charcoal powdered in to increase germination rates.

Yes, MSS = multi spore syringe

A sporeprint which is two years old, is very old and often give alot of problems germinating. Letting the spores "soak" a couple of days in a syringe might hydrate them enough that they can germinate.

I only know charcoal to be used as coloring. Didn't know that it could help with germination :shrug:

I've had year old prints I got from other members take over a month or 2 to germinate. How long have you been waiting? Also charcoal sometimes can cause issues so stay clear of it. Stay away from any food color as well, which I see you did, until you get good growth. It's purely cosmetic and when the myc eats the color it may cause confusion if it's contaminated or not.
I've also had some that just never germinated even after 6 months.

Those were the two plates I have put into the fridge.  Potato flakes, a dash of yeast and pinch of sugar.  I meant to put just a drop in centers but squirted a bit.  Supposed to be APE cupensis jars of WBS are almost ready to spawn, I hope. Three and a half weeks for the jars, I fridged the plates about a week ago.

Think there are good transfer points?, and should I use a needle or cut out a piece??

Unfortunately you are looking through the Agar, which is fairly cloudy, lit from below.  Tops are impossible to photo though with cheap phone...

can’t go wrong with needle biopsy, but usually with plates like that i just cut a big ol chunk & slap it on a new plate

Ok, good to know they are good enough to use that way...  The thinner one started to peel away a bit, which was why I wanted to transfer fairly soon.  I have three pasty plates in 1/2 pints Im waiting for condensation to disperse.

i didnt think a few years was really that old considering some mush heads germinate 20 year old spores, i even have garden seds 5 plus years old that germinate fine ( i know spores are different )


This is kind of a curve ball for me I had no clue just a couple year old spores would be such a bitch to germ. Guess ill have to get into some syringes, any good write ups can point me toward?

These plates have been going just over 2 weeks, im guna keep my eye on them and hopefully they pop. There is puddles of water where the spores are so im sure they will go just may take a while.

Ya im not into additive into my agar, straight MEA for me.

I don't see the problem with those plates, other than being slow. If my ayes aren't playing tricks on me, I see plenty of action. Got a closeup of bottom right?

Odd 2 plates are turning a brown color, but I see "normal" growth. Can someone please help this noob out with what is actually happening and if the two are fucked?

Just sterilize a shotglass, and scratch some spores into the glass in your SAB.

Good syringe tek for MSS and then just fill the glass with the spores with the water from the syringe, suck it up and press it back into the glass. repeat 2-3 times and then just keep it in the syringe and put the needle cap back on.

They way I would recommend. But there are many different ways, find it works best for me. Maybe someone has a better Idea.
