Thanks! Brf? Haven't seen this use same amount as you would potato flakes?  I'm just trying different agar recipes haven't liked pda agar but haven't tried the stick blender yet.  Maybe I'll do that next. New to agar have done about 150 pda plates with lots of tomentose growth not much rhizo so just running as much as I can just to learn more mostly but screwing up though.

So I was given this plate of what was labeled "Albino B+".
I've actually never heard of albino b+ or even Leucistic B+ :shrug:
So I'm thinking theres a chance it's a mislabeled culture... idk. Anyone heard of Albino B+?

Anyway, it arrived lookin pretty bad, I've had a bitch of a time cleaning up 4 lots of transfers finally have something decent to work with.

Had a look at the original plate today and there are a few pins popping up.
Is there any point cloning some pin tissue now I've got some clean culture or should I just do it anyway?

Wildaussie I do 6 gram brf 9 grams agar and 450-500 ml.

Does anyone know if its ok to use clothes dye in agar. I didnt know charcoal wouldnt fully dissolve and i wanna make a bunch of black plates.

Haha, yeah... I abducted the big pin on the way to the party, threw it in the back of the car and performed surgery on it. Stole its organs, put em on a plate of agar and sent it back a few dimensions :hehehe:

So I did do a biopsy on the big pin, cut it down the guts, slapped a bit of either side on a plate each.
Figured what the hell. If it fruits well then we're in action.

Still not sure about albino b+ though.. :shrug:

The growth appeared to be normal at first.
Rhizomorphic ropes/tendrils have begun to grow inside of the culture, near the original transfer.

Could contamination be causing the newer inner ropes to try to grow over the older outer ropes?
Could this type of mycelial growth be indicative of a vigorous culture?

I’d guess something hidden.

Look at the top of your transfer. The growth is really grainy and fuzzy not cube like.

Suspect to the fullest or I’m wrong but I don’t think so:noway2:

I took a dried-up pin from a 9-month-old pf puck that was on my shelf that I forgot about.

This tidal wave mycelium is crazy fast.

I've sent plenty of cultures that looked like that to wonderful success. I don't see any cause for alarm, just a culture that is deciding to group up into different colonies with differing desires and expressions and goals. They obviously love each other dearly and are reluctant to part.

A little while ago I sent a note to the poster saying "I think it's OK but I don't want to contradict a TC on board when I'm not sure."

But if Nichrome says it looks OK, I'll say it. I'd transfer some of that, but also grow this out and send some to grain.

Another point, I'm not sure if Inf Monks was asking about this too, but I think some of the concentric circles are just from temperature variations.

Every time you bump or move a plate, warm/cool it (:whathesaid:), change light intensity/spectrum, exchange air, alter internal humidity, or change the environment in any way, a new ring can form. It is just new growth direction. It is cool to watch the aerial mycelium in dish vibrate to the sound of your voice nearby or music playing. They are very sensory perceptive organisms and adapt on a minute to minute basis to environmental changes.

I'm sure many of you have noticed that taking a wedge or cut from a leading edge will often cause variation in growth pattern and or a ring to form.

Opening them up brings in fresh air, rich with brand new oxygen. A fresh supply of oxygen allows for faster metabolism.

Sometimes opening up the plate and exchanging the air can trigger pinning too. A neat trick if you hunt plate pins or want to see traits in an unknown culture like cap color or shape.

is there a basis for hunting plate pins or is it best to just clone a bunch of stuff, i wanna make a bunch of pasty plates cus the myc can breathe. i have some plates similar to those of mr.aliens but water always pools in those and im always paranoid to loosen them up.

If I get water in plates (from condensation usually) I put the plate  upside down for a few days or a week. The plate will still grow and often the water evaporates back into the plate air, or just doesn't disturb the mycelium. Water rolling on plate can spread any tiny contam everywhere.

With plins is the same as any cloning, you want to pick the ones that are the most interesting, fattest, tallest, first to grow, fastest to grow, or any combination of the above.

The only culture I grow lately is a clone from a year ago, was the first plin in a plate of x7x.

If you want to try to force plins, mix your agar with a little extra water so it won't dry out. Plins I've seen form any time around 6 weeks give or take a week or two (your mileage may vary), but sometimes plates dry out before then. You can also seal the plate with non-porous tape, like scotch tape. Will prevent drying out.  Per Bod, sealed plates have enough oxygen to grow.

Different than what Nichrome said, but it's not contradictory that more oxygen might have same effect as lack of it: making fruits.

Fruits form when culture feels "This is it. I HAVE to throw spores now, this may be my last chance. I must survie and carry on."

Same reason bacterial grows often fruit, and sometimes do it quick. (Though I don't for the life of me know why people intentionally send bacterial spawn to tubs, unless it's something rare, like some wild shrooms you printed.)

Also the same reason more baby humans are born 9 months after a natural disaster or war attack. haha.

Plins usually form when the plate is entirely colonized, same as with shoeboxes. When they run out of food, they throw up fruits to try to spread spores.

Take clones of plins before they open caps, and cut off with scalpel (flame sterilized, of course) above the bottom to avoid getting non-clone mycelium. Then just push the plin into a new plate with scalpel (flame sterilized, of course)
