That cutter looks nifty. I'll need to pick one of those up.

Nasty.. what are those, mites?

Yeah, if I switch to parafilm, I probably get the one with the cutter case.. it's a few dollars more, but it's refillable and looks like it would be convenient. Plus the hard plastic will help keep the roll from getting damaged...

Like LotKid said, sometimes you've gotta pay a little  more for quality equipment.. it really just depends on preference and how much you want to invest in the hobby

I'll probably stick with my dollar store wrap until I get my flow hood built and then upgrade to parafilm sometime after that

Ah, so grafting tape is the same thing? I didn't know this, I will have to reach out to someone that asked if grafting tape they found on Amazon meant for plants was a suitable substitute for parfilm or Saran Wrap. Thanks Nichrome

:themoreyouknow:

I am a little embarrassed to admit that I have always used Saran Wrap rolls cut to size because it was what I did when I first started growing years ago and it wasn't broke so I didn't fix it. I think it's finally time to step up to parafilm.

There is a skillset/learning curve with any new tool. Bear in mind.

Rot, yes those are mites and they exist everywhere. Even on orbital space craft. NASA has problems with them.

A lot of the time when tubs fail (mold, bacteria, etc) these are the culprit too. They are microscopic.

I use cling film also

and here I was recently thinking, how about I just wrap plates once around, would save a fair bit of time on a large stack of plates

then I saw that picture of the mite

That photo is inside a colonized dish. You can see all the eggs bound up in the mycelium that has been all chewed up.

This is healthy mycelium at the same magnification.

Very cool to actually see what they look like..

That makes a lot of sense... I unfortunately had a fruit fly problem pop up at the beginning of spring.. and the couple tubs that they got into contaminated shortly after.. it's one of those "chicken and the egg" situations, but if I were to guess I'd say that they played a big part in the contamination by carrying some sort of spores and transferring them into the tubs. Especially considering that 2 of the tubs were pf tek shredded to bulk, so I was pretty confident in my spawn on those tubs.

I can only imagine the sort of crap that mites can carry with them.

Fruit flys, or fungus gnats? Fruit flies have a rounded silhouette like a smaller version of the common house fly. Fungus gnats feature dangling legs and long bodies that make them appear similar to a small mosquito. I'm guessing it was fungus gnats.

I'm pretty sure they're fruit flies because they originated in my kitchen and moved their way back to my grow area, but I could be wrong...

I ended up making a crap-ton of traps using apple cider vinegar, hanging fly tape, switching to fruiting in bags, and storing fruit differentlyin my kitchen... they're finally all gone.

If they return, I'll try and see if they were actually fruit flies or fungus gnats. Hopefully with all the new precautions I'm taking around the house they will stay gone 🤞

My poor cat jumped into one of the hanging straps of fly tape  thankfully other people have had the same thing happen, so a quick Google search told me that wiping her fur with a paper towel that had vegetable oil on it would get all the sticky shit off of her.

First go at using a spore print from a recent flush. I was surprised at the amount of growth already. Either this is all bacteria, or are they simply new hydrated spores ready to go. I scraped a lot of spores off of the print with flamed loop cooled in agar. Probably too many in hindsight.

My question is, if this is good mycelium (is it? Hard for me to tell at this stage) should I be transferring these isolated colonies at the end of the streak before they merge with one another?

Looks like mycelium to me

It's all preference.. some people prefer to transfer from germ plates quickly before comtans get a chance to take hold, others prefer to let them grow out a bit.. I'd probably take some transfers and then keep the plate and let it grow out

It looks good.

I'd let it grow out 2 to 5 days so you're only transferring mycelium, not spores. And maybe you can see some mycelium performing better than other, and pick the more robust mycelium.

If you get contam I'd transfer then, away from the contam. if you want to post another photo in a few days we can give opinions on where to transfer from.

No need to transferring the isolated colonies before they merge with one another.

A 4" roll of parafilm will roll 1500 plates when you use a 1"x4" section, which is just right IMO.

You can do more if you are anal and careful but a 1"x4" piece works very good and is very easy to cut off the 4" roll. Look like dis

I recently bought some lab grade markers that don’t come off with iso. I wipe my dishes before opening them or putting them in the sterile air in front of the flow hood too.

I’m a noob and super paranoid about contam.
