First, I'd recommend next pix you take, don't have your hands underneath. The blue gloves make it harder to see what's going on. Just lay the plates down on that neutral dark background.

I'm going to ask for someone else's opinion on these. They look like OK photos, more or less in focus, but I'm having trouble seeing what's going on in the ones that have small dots.

Are the small dots transfers, or where you touched a swab?

Is the big circle a transfer that grew out? The big one looks like it might have some rhizo mycelium worth transferring near the top outer edges, especially around 3 oclock. but may have some light contam colonies off shore around 12 oclock.

Inside of that one I'm having trouble telling if it's fluffy mycelium or mold.

The first pic, and the bottom two pix look like they have some wet bacteria near the bottom on the bottom two, and all on the left in the first one. 

Are these plates sealed? I don't see tape, but they look like seal was on then removed.

Anyone?

Don't use heating pad, and you're running them too hot. 78f will almost guarantee contams, especially at first. Run them at room temp. If warm enough for you, is warm enough for them.

Syringe to plate often leads to contams, but sometimes can be transferred away from.

Better is swab to plate, even better is spore print to plate.

Is your DIY flow hood a box fan and a cheap filter? Those don't work. Better off making an SAB"

Costs about 10 dollars. Use the biggest tote / tub you can. I used a 107 quart tote. Smaller will be a hassle. Here's a good tek on making and using one. Though skip the mentioned chlorine, is bleach, you don't want or need it. If you use a hot coffee can to make the holes, wear protective gloves to prevent burning your hand. Or hold it with pliers, and do it outside to avoid breathing fumes of plastic melting.

All came from MSS… on the one with all the dots I used too much so I tilted the plate a few times to spread it out. I got a SAB(120qt), used it to take the pics. Imma chill out on the box fan, but I thought my shït was on point tho🤣🤣🤣.

I try to keep my Sab sessions short so yes Ill leave it lit the whole time. If I have lots of work to do Ill turn it off occasionally so the touch can cool down.

Well, cling wrap is meant to stop air flow in order to preserve food. Parafilm allows for gas exchange

Parafilm is a great and useful stuffs for so many things. That's why every lab supplier sells it to nearly every lab.

Not like labs can't get cling wrap. That's in every lab too but not usually used for what parafilm is for.

We all have preferences. None of them are "wrong". There is no cheating in art.

Hmm didn’t know that I thought they were used interchangeably. Perhaps I’ll try parafilm after as well…thanks all.

Anyone know approximately how many plates you can wrap with a roll of parafilm?

I noticed they have a refillable dispenser with a cutter,. Thought some of you might find that relevant since cutting methods were being discussed a few pages back

Length of the roll in inches will tell you. If you use one inch per plate then you can wrap 69 plates from 69 inches. If you use half an inch to wrap plates then you can wrap 420 plates with 210 inches of parafilm.

It depends on the size and the wrap you are doing.

For example I wrap poured plates with two squares (folded over lengthwise) just to store them before usage. I double that when I expect a plate to sit a while with a culture in it. 4 squares folded lengthwise. That keeps bugs out and makes sure there is no possibility of micro tears or holes and allows for handling without having to be so feather fingered.

These fuckers can chew through one layer of film or cling.


That cutter looks nifty. I'll need to pick one of those up.

If each square is 2 inches and you use an average length of 3 squares that's 6 inches average per plate. A roll of parafilm is 250 feet. 250 x 12 = 3000. each roll is 3000 inches. 3000/6 = 500 plates wrapped average. 1 roll per case of 500 dishes. :super:

I like the double rolls. You get more film for your $ and having a wider film is advantageous for adaptability although it requires more cutting. I find the paper layer to be helpful in a lot of ways as well. If you don't like the paper you can get grafting tape rolls. It is nearly identical although quality control is crap with that stuff but it is cheaper and has no paper.
