500ml water. 10g agar. Mix. PC. Put culture on it. The culture kinda invisibly expands. You only really see it if you hold it towards light. Transfer. :laugh:

I mean you should be able to see the mycelium.
It continues to use the agar from the original transfer and grow outwards, and nothing else will grow on the new agar because there is no nutrients for it to colonize, really the mycelium isn't necessarily colonizing water agar, in a way, as there's no nutrients for it to use, it's more or less reaching for nutrients, almost like a plant stretching for light.

Very good analogy. I understand. Can't wait to try it and see how the mycelium looks when reaching across the plate.

One thing I’ve noticed as far as concentric growth is that if you take a wedge from the leading edge, a lot of times it’ll grow slower on one end. If you take the wedge further down where it’s been totally colonized that doesn’t seem to happen.

Also, when taking wedges, you can accidentally kinda tear one side, whilst a different side will be a clean cut. Different jump off times then.

I like a bent stainless steel drinking straw for an agar punch.  The ones I use are on the smaller size and give me a 5mm diameter sample.  Thin metal, so they heat up and cool down quickly. About a dollar or two each in a 6-pack in bigger grocery stores, TJ Maxx, Homegoods type of places, or of course Amazon.

It helps to sharpen the ID of straw with a deburring bit, but it's not necessary.  I have a few available for a session in case the punched-out agar bit gets stuck in the tube, which happens occasionally.

For smaller samples, I occasionally use a 10 gauge luer-lock needle, for 3mm discs.

Man what the fuck. My custom straw punch looked exactly like this. With the tiny orange handle and everything :laugh:. I tried to sharpen it one day and messed it up though.

Pro tip for punches: rotate the punch a smidge when you are inserting into the agar. The cut is 100% more clean and beautiful. Plus less of a shake, so less of a chance of anything that was stuck on punch getting lodged into the 'gar

Every once in a while if the straw cools off too much before the punch.

Could I get some opinions on these please?

First dish was an aerial transfer from a germinated dish which was obviously contam - is any of it good? it looks pretty weird to me:



Second dish was another aerial transfer from a germinated dish which also didn't look too hot - the color is concerning me?



Finally I had an LM culture knocking about (which I didn't manage to grow on grain, and switched in agar in part to find out why) - I'm assuming this is nothing good:



any help appreciated!

Once in a while.  I keep a few spares around so I don't have to stop and clean it, but I blow it out a can of office duster when I have a chance. (or my air compressor but that overkill)

First ones wierd, going to need a couple more transfers probably. Hjenstly I've found with those feathery looking cultures they're usually ass also, kind of like scalloped cultures, you might be able to clean and fruit but they usually suck if they don't turn around after a couple transfers.
The second one looks better but makes me wonder the strain and your agar recipe?
LM looks fine, and it actually looks really good, unless it turns absolutely green in the next couple days ha. Looks kinda fluffy for LM but looks good. I'm guessing your transfer was rolling around or something. Looks like an interesting LM strain even, only a couple LM strains I've had forms mycelium like that in the beginning. Unless maybe your colonizing your agar below 69F maybe, in those cases I have seen fluffier LM growth more commonly.
