Swabs, are great. But I wish i could inoculate more than just a dozen few plates with it. considering thr thousands of spores on it. I wish i could reuse or save a portion of it.
Well maybe that's where it started. When I tried to saran wrap a quarter of a swab remnants after a partial use. Or maybe when i was looking into some breeding info.
Anyway before we get started i wanted to point out that there is indeed a very similar, and identicle in parts, write up on this already, as far as making swabs syringes. 1. So lets just get to the point. Need a swab.

2. Need foil, syringe, jar.

3. Put 10 cc water into jar. cover jar and syringe with foil.
4. PC 15 minutes.
5.Remove items in front of flow hood before pressure reaches zero, or move items to SAB, wipe everything off with Iso.
6. Cut the tip of the swab off into the jar in one quick motion.
7.Shake and let sit for 5 minutes, repeat for 30 minutes. I did this while doing other work.
After this the swab should be relieved of all spores, and the swab itself should have nearly "dissolved" into the water.
8. Now unwrap your syringe and suck it up! Recap and your good to go!
Now rather than taking that swab and smearing it over 8 plates, or ripping off little pieces of it, you can simply get some agar, and put 1 drop from the syringe on it, after shaking vigorously of course. and idk how many drops are in 10cc but I bet you can make at least 100 plates. bet? or you can just make 1, and save the rest!! you could also put a few drops on a sterile surface and use a inoculation loop to get even more out of it, or perform a serial dilution for a number of reasons. Your sterile technique is valuable, but these will need to be cleaned up just like any other spores.
Looking closely you can see the fibers from the swab in the syringe, although no spores to the naked eye.
  and not only is this an albino strain, but you cant see a spore. you cant see two spores, idk how many you would have to clump together to be able to see with the naked eye, but my guess is there's over 100,000 on the tip of a swab. and you need 2 spores to germinate for agar.
if you want to try and breed something mix spores together, make a swab from 2 different strains, the choice is yours.
Store these syringes in the fridge for years.
BONUS
Ever buy a syringe of your favorite strain and things happen and you have to buy it again? Isn't that frustrating? How many times? Ever realize there's probably millions of spores in a syringe (at least from Sporeworks.com), and you only need 2 when your inoculation? Ever thing well maybe i could stretch this out a bit?
Well you can. Pretty much the same exact way.
1 difference.
Instead of cutting the tip of the swab into the sterile water, get your favorite spore syringe
 see that big ole booger in there. Seriously enough for like 1000 plates, who knows ha.
were going to shake the syringe up nice and good so we don't take the whole booger.
and were going to put about a single CC out of the commercially prepared syringe (maybe less or more depending on the concentration of the original syringe), put it into our sterilized water, shake it up and suck it up in another syringe (no need to wait and repeat like the swab). and just like that bam you have 2 syringes. And if your doing agar your probably only needed 1 but now you got one to send to a friend!
Ide suggest understanding serial dilution to some extent if you really want to appreciate these concepts to the fullest extent.

When you say cut the tip off, do you mean below the cotton or just the very end of the cotton where the most spores are?

I’m in. I have a couple swabs (pairs) that didn’t do jack on agar. Going to try this with the left over swab and see what comes of it.

I will say that if you leave the syringe in room temp, the spores will use the cotton (if that’s the swabs you’re using) as a nute source and germinate in the syringe barrel.. same with any contams that are in with them. Just a head up there
