With petri dish lids, what is good sterile technique when working in an SAB?

My process looks like this

  • Flame scalpel
  • Lift lid of donor petri dish + hold lid above dish with left hand
  • Cut a wedge with right hand
  • Place lid back on petri
  • Flame scalpel
  • Lift lid of donor petri dish + hold lid above dish with left hand
  • Stab the wedge with right hand
  • Place lid back on petri
  • Lift lid of receiving petri dish + hold lid above dish with left hand
  • Place wedge on receiving dish with right hand
  • Place lid back on petri

The process is somewhat frustrating because it is not easy to cut a wedge using one hand while holding the petri lid with the other hand.

Would it be risky to place the lid down somewhere in the SAB and hold the dish with one and and cut a wedge with the other hand?

Where could I place the lid and should it be face up or face down?

Use the petri lid to hold the petri while cutting, replace the lid once cut and move the transfer to the new dish using the lid from the new dish to hold down the receiving dish again.
Only open the dish enough to pass the transfer through.

Thanks for your response.

Do you mean place the petri dish into the petri lid (upside down) while cutting?

I used to set the lids face down on an iso soaked paper towel in the SAB.

It’s definitely better to learn how to hold the plate and lid in place but it’s not horrible to take it off for a second if your technique is solid.

I ended up getting a piece of plastic sheet and cutting a hole just big enough to slide a plate into so it couldn’t move. Made things way easier.

You do the donor plate first,  one hand to cut and the other to use the petri lid to hold the lower dish down.
Then close the donor plate and open the new plate using the lid again to hold the receiving dish.

I have to work on my SAB technique. All of my pasty plates are contaminated except the control (the one I didn't use).

I tried the lid face down on an iso soaked paper towel method.

I'll probably go back to using a hole punch. It's annoying to flame it but it makes the process of cutting and transferring wedges quick and efficient.

I ran out of grafting tape and decided to use glad cling wrap.

I bought a big roll of the cheap in house brand wrap and a small roll of the glad branded wrap.

It's astonishing, it works well! The in house brand wrap is dog shit but the glad wrap is fantastic.

Same pics, same question - any ideas why my mycelium is only growing clear? These pics were from a couple days ago and they've now frown out even further, still completely clear. If they don't fill in in a day or two I'm going to have to take invisible transfers

I used D3_Myc (formerly D3monic) agar TEK and recipe, which is what I've always used and never had this issue.

Here's a picture of my dog as a peace offering for my double post

Not sure if you're directing that at me mate?

Any other agar tips you've been trying to tell folks for years?

You have to understand, for a rookie, there is an overwhelming amount of information on this website to sift through.

With the proliferation of concepts like "Uncle bens TEK", some ideas aren't given the reverence they deserve. There's a lesson in here somewhere for me :P

cut wedge, stab and remove. there is no reason to flame the scalpel between the cut and stab.

It was, that's what the "re: username" deal means.
I fully understand the overwhelming amount of info available for noobs to sift through.
I was one once and sifted through almost all of it.
Using the search function properly helps.
Also asking specific questions in active threads full of knowledgeable folks (like this one).

I posted this in "new tools of the trade" a few month ago and still using it with great success for transfers.

It's a 16g or sometimes 18g needle stuck to a nail. Takes only a second to flame and doesn't need to be cooled in the receiving dish.

Glad I made the switch to cling wrap. I got some hostess brand rolls from the bargain outlet and it's pretty good. I need a dedicated cutting tool that wont pinch the wrap causing it to stick at the edges when using.

Some say a pipe cutter is good. Not a fan of knifing.

LotKid is gone? Been a long year.

Mold or gold?

Only one of my plates that I streaked with an APE swab showed anything after a week. Is it what I want or is it some other crap?

These swabs are always so damn weak. If I take a spore print and rub a sterile cotton swab over it I can easily streak ten plates or so with it. Ten plates that will go completely crazy withing 3-5 days.

That's what I was questioning, haha. I miss context sometimes and thought that was aimed at me.

That was a misunderstanding on my part.

I understand. I miss a friend too.

Agreed btw. That's why I'm here asking questions :P.

I used a serrated knife then sliced the spiky bits off with a scalpel.

Haven't heard of a pipe cutter before, will look into that.

Isolating it more and more just out of curiosity.

Advanced search is here:
https://www.shroomery.org/forums/search.php

I limit it to searching only from the last 5 years. A lot before that isn't the best info anymore.

Also tick the box
"Only show one result per thread"
or you'll get too many results from the same thread.

If you get too many results, MAYBE only search for posts by Trusted Cultivators. Though there are solid non-TCs. You'll start to notice after a while. Often (but not always), people with good replies have a lot of General Reviews (click the shrooms above their avatar over on the left.)

If still to many results, maybe limit to last 3 years.

Hey all. I have a newbie question. I made 200ml of grain soak agar a couple weeks ago. I poured three plates while it was still molten my from the PC, then closed it, but I didn’t store it in the fridge. It has just been on a shelf. It has since solidified and has a bit of liquid on top. Is it good to re-melt in the microwave and pour more plates, or do I need to start over.

Also, can I melt it without venting the lid? I don’t want to introduce contams.
