These are the best I could get - it's a pretty tricky picture to take because they only show up when the light hits them head on but then my phone blocks the light and they disappear. The speckles look like hard water stains on glass, but obviously aren't lol.

Yea I've gotten that from time to time. I have been meaning to get some kind of microscope to check it out. The plates seem to work fine.

I think it's either "nothing/physical phenomenon" (ionic bullshit?) OR yeast of some kind of your skin.

Whatever it is doesnt ever seem to grow into anything more than the little nearly invisible spots.

I've checked em with a scope. That's how I figured what they were. Plus, if it were something else, the plates wouldn't do fine.

I'm thinking some kind of ionic bullshit then

Quick question peeps, I tried the search engine and google but am getting mixed info

How long do you let your fresh agar plates rest/dry for before inoculation? I'm reading some pretty conflicting things. What do you all look for to tell when they're optimum to inoc?

Cheers :hatsoff:

You can impregnate them as soon as they harden. If you've poured hundreds and have great success rates, it's safe enough to do. Even skilled agar pourers accidently shit in a plate here and there. Giving them a week or two to chill is good because you'll allow the shit to bloom.... If it's there. I try to pour new plates before I'm out of old plates for that reason but I'll pour and use immediately if I have to.

I been using my slow cooker for agar bottles. Keeps em gelled as long as you need.

Just ordered the gellan gum I mentioned previously, I’ll be testing and posting results.

Also ordered potato flakes, dex and charcoal to test my experience with those. Not particularly interested in any of those other than the gellan but it gives me something to do.

yo G that slow cooker is fucking brilliant. just in time for agar orgy day.

these LC tests...fucking nasty, nasty, or go to grain from the LC? I assume the penicillium landed during streaking.

I'm considering buying a box of 60mm plates.

1400 x 60mm plates is about $200
versus
500 x 90mm plates for $100

I'm finding it hard to visualize their true size.

Does anyone have experience with the 60mm plates?

One 500ml media bottle makes about 20 of the 90mm plates for me (with some left over).

Presumably a 500ml media bottle would pour more than 20 60mm plates.

I generally don't let a culture grow to even 2/3rd the size of a 90mm plate before transferring so maybe 60mm would be ideal.

60mm = 2.36" which is like the size of my sunglasses lenses. Way too small of an agar plate for my liking

60s would be ok for knocking up jars and early transfers where you just let it barely grow before doing another one.

Stuff like germ plates or iffy cultures it’s way better to have 100s so there’s plenty of room.

60mm is roughly the size of the holy grail no pour plates. It is honestly workable but bigger would be ideal in certain circumstances.
You can also put 2 or 3 transfers in 90 or 100mm plates if you don't intend to let them grow out.

Thanks mate, that's a great visual representation of their size.

Yeah guess I'll stick with the 90s for now.

I'm running so many different cultures on agar right now, it has become hard to manage.

It looks like 60mm plates would probably make that even harder to manage, lol.
