Plates, plates, plates! (I need practice taking pictures of the damn things. I need to make a photo box for plates or something. ANYWAYS.)

These are pretty much all tissue transfers done in a SAB with either flamed tweezers or a scalpel. MEA (probably on the higher nutes side).

I'd love for some of the "BM Chonger" plates to make it -- alongside the MDKs and the GTs of course :wink:.

Here are a few (there are more) that I'm considering transferring sooner than later! Any and all advice is *welcome* and appreciated -- this is my first time with anything except bacteria on agar.

How long does everyone let their plates sit before using/wrapping/storing them? I usually go 24 hours but I need my office soon and I've got my SAB set up right in the middle of my desk lol. Didn't think that one through...

I been using my slow cooker for agar bottles. Keeps em gelled as long as you need.

Just ordered the gellan gum I mentioned previously, I’ll be testing and posting results.

Also ordered potato flakes, dex and charcoal to test my experience with those. Not particularly interested in any of those other than the gellan but it gives me something to do.

As soon as they've solidified. Depends on the temp they were poured, how high they are stacked (middle plates take the longest) and ambient room temp. I'd say an hour or so should easily do. If in a hurry and curious, just carefully extract the middle most plate from the biggest stack and see if she's flaccid or aroused.

That's a lot of tissue samples. Take it from ol wall o plates himself, it is possible to bite off more than one can chew and end up accomplishing less than you would have if you had kept it a little simpler. You do you though. OK........ here we go

Pic one: Looks like it may be clean or may be fighting something. I see no particular place that looks better than any other. Just transfer and see what happens.

Pic two: Looks to be certainly fighting something. I'd just let her keep growing and see if anything good looking appears.

Pic three: Not the best pic but I'd choose the lower plate and leave the upper to chill and grow out. Bottom plate looks hopeful. I'd transfer from 11:30 - 11:45.

Pic four:
Top: Looks hopeful. I'd probably snatch up the 8 o'clock portion.
Second row:
    Left culture: Probably be OK taking from around 2  o'clock or 6 o'clock on the right growth.
    Right culture: Meh. Seems quite fishy. Might get something out of the 1 or 4 positions. PS, are you cloning from gill tissue?
Third row:
    Left culture: Looks pretty good. I'd snatch at 8 o'clock.
    Right culture: Meh. Looks like trash.

Pic five:
      Top left and bottom right cultures look most hopeful. The pics are out of focus so it's really hard to say too much. I think I'd snatch 9 o'clock from the top left culture and 5 o'clock from the bottom right culture. PS, confirmed cloning from gill tissue. This results in the germination of shit tons of spores and negates the main points of cloning.

Pic 6: Bad pic. I can't tell.

Pic 7: 4 - 6 o'clock look pretty hopeful.

Pic 8: I'd let her grow and show her true colors for a couple days

Pic 9: Looks pretty hopeful. Still a bit early to tell but you could probably snatch up something useful from almost anywhere. I think I'd let her grow as well.

Dually noted about the gill tissue. I actually suspected as much (and had already taken some tissue from the stipes). I’ve also accepted that I’ve taken on a bit much. Simplifying things seems good. Transfers shall be taken. Updates in a few.

(edit) update: In efforts to simplify, I took the best and removed the gill tissue plates and any showing signs of ick to the we'll see later stack. I'm going to let these go until at least tomorrow morning before making any transfers.

Just finished up about an hour in the SAB taking nice transfers and wrapping my newly poured plates. As I'm packing up I held a plate up to the light to see how clear I got em and EVERY. SINGLE. FUCKING. PLATE has 1/16"-1/8" speckles all over it..! Are there even any contaminants that would germinate in 15 hours? I've only ever had like 3 plates contam out of ~500 plates. WTF!

All the speckles look the same and a large majority of them seem to be surrounding an air bubble/divot/imperfection on the plate so I'm really hoping it's the result of pouring agar that is very close to gel and that I didn't contaminate 2 sleeves and waste all my t-2s.

These are the best I could get - it's a pretty tricky picture to take because they only show up when the light hits them head on but then my phone blocks the light and they disappear. The speckles look like hard water stains on glass, but obviously aren't lol.

Those will do. Tis my belief that you are seeing minute variances in the agar surface that are catching light in a different way than the flat portions. I've seen similar from time to time. Dead give away is that it's only happening around the bubbles. Makes sense that the bubbles would cause curvature of the surface. Make sense?

I do a half spoonful. My best estimate is 1g per 100ml but adjust accordingly.

I went from having no ape cultures to 7 different varieties thanks to amazing friends. Excited to do some comparison grows. The black #1 is one I’m excited to potentially use in some crosses.

Cheers D3. Half a US tablespoon is 7.39g so I’ll try different amounts around this.
