Very informative write up!! Thanks so much. I've got a batch of transfers I need to do in the next couple days, so this couldn't have been more timely. It will help me make much better informed decisions on transfer site selection.

Very helpful article!

I've been working with Agar and accidentally left a flask with Agar open to the air in the room I do my work. I noticed quite a few growths that were white and would be difficult to distinguish from exotics mycelium, and I'm wondering

How does one ensure the culture isn't mixed with other white mycelium and if it is, how does one separate them?

I'm thinking maybe I need to make a special dish or maybe transfer to a slide and view under a microscope to look for spores (indicating lower fungi)...?

Then, if I can't find a clean sector, how can you separate them? I've heard pour Agar over it and only the higher fungus will survive/grow through the Agar, but I tried this with P. cinct mycelium and it seems to have died.

Great post but there are some very wrong parts in the middle (the part on "sectors" and "monocultures") that time has proved wrong.

I tried to contact Stro to see if he'd correct it, but he didn't reply and hasn't logged in for 7 years (since 2018).

So I copied his post and re-posted an edited version, with credit, here:

I don't have time to compare the two, but im guessing something about anastomosis causing sectoring to be an unlikely or even impossible rout to a monoculture?

sectoring could be caused by a number of things that u wouldnt be able to tell by visuals alone

i do agree with the notion that trying to isolate a "monoculture" thru just transferring sectors is probably not the way to go about it

but i dont think saying that bc something sectors that it 100% isnt a true isolated monoculture, the fact is u wouldnt know anyway

theres gourmet mushroom papers about commercial production strains that seem to suggest degradation of the culture was the cause of sectoring or that it was thought to be the case

Mutation also happens spontaneously, and could cause sectoring
