Re: Stro's Cleaning and Isolating On Agar [Re: rudimentary]
    #26747078 -

Curious if anyone knows how to get myc on agar to form rhizomorphic growth.  I'm well aware that it can always go back and forth, and that there are great strains that are tomentose, but for my purposes I often find that rhizomorphic allows me to identify sectors easier more often than tomentose.  Wanted to see if anyone has any input on the matter.  I find that if I take an isolate or mycelium with multiple genetics that is on a piece of grain, and transfer the grain to the agar, I often (roughly 90% of time) will get rhizomorphic growth.  If I go from agar to agar, it seems tomentose around 80% and 20% rhizo.  Anyone think perhaps more complex food sources have something to do with this?  Most of my agar preps are PDA or MEA - I've tried fiddling around with nutrient density in those dishes, but doesn't seem to have much effect for me.

For me thinly poured plates with like 4g lme to 400-500ml does it most of the time.

That is super thin. Like SSLI thin if your doing 4 g of LME.

I usually do 6-7 LME 10agar.

Iv never had tomentose growth on agar only rizomorphic I clone from fruits around the base of the stem I get rizomorphic off the first dish an easy can see sectors

thank you for the write-up Stro!  this helped me a ton in learning how to see the sectors and where to transfer from, among a bunch of other little details.  nice and simply explained. again, thank you!

Thank you for this Stro!

I read about 'strain isolation' on agar in a bunch of places
but the process only became really clear after your post.

what a great post.
I plan to do this with a spore syringe since my first mono tub was a fail.
My question is how long can you keep an agar plate thats inoculated  before placing in say a spawn bag.  Also when do you know it is ready to go into a spawn bag.
Also can you keep dish C for a while stored in the fridge or dark space Before using to inoculate.

I plan to buy some pre-made agar dishes on amazon since I don't have a PC and for a first time seems cheaper to get them rather than all the stuff I would need plus time.

If there is a good write up on next step l would love to see it. I know there is a lot of good stuff on here just overwhelming sometimes and I seem to go down too many rabbit holes ha!

They were however moved to a higher temperature for colonization.  There appearances speak for themselves. The colonization times were faster and characteristics are distinctive. If I don't keep medium and conditions consistent, the mycelium tricks me up.

tnx for great info, well done. but here I don't get this. how mycelium trick you? you mean if temperature is not consistent the pattern would be different and they can appear as sector or contamination?

Thanks for this! Very informative! Ive been struggling with figuring out what to transfer in order to clean up my sample, this definitely gave me an "ah-ha" moment.
