What do you guys think of plastic Petri dishes already filled with agar and sterile like they use in medical labs? I found a place to get them. Didn’t know if they would have the same nutrients needed for myc. Any thoughts? Sleeve of 20 already ready to go sounds Good to me.
And if I have a laminar flow hood, how important is the “lab”. Won’t the work done under the hood be good and clean ( provided it’s done right).? Ie, I don’t need to hyper clean everything since work will be done under the hood?

no pour in a sab is what i do pretty easy and cheap

 Not bad for a germ plate especially from a swab. The satellites look funky in the pic but I think they're clean colonies in 1st person.
Another killer germ plate from Spinlymass, thanks bro.

I am new to LME agar. The first attempt worked well, the second I cracked the lid when their was pressure and it sucked in contams, made me want to cry as I didn’t give it too much thought and a few days later every damn plate was ruined. This time I thought I was so careful but all my plates look messed. Is it the agar? Is it contams? Is it ok? I was going to throw them out when I saw the weird swirly stuff but decided to watch for a bit to see what happens.
This one I didn’t germinate.

This one was a transfer.

They all pretty much look like that. Is it just sediment? Need to go back to potato or GW lol

Well I just got my first isolate of Orissa India, I think I did 4 clones to agar and 3 have obvious sectoring but the fourth is a perfect circle. I transferred to two more plates so hopefully I can use it by next week. Will post pics when I'm home.

If it's not moving around or growing I would say it's just sediment...it happens

Might be caramelized sugar, how long did you pc? Over 20-25 minutes makes it happen to me. It doesn't hurt anything. If you put food coloring in, 10 drops or more per liter, it's easier to see some contam stuff and you can focus on what's on the agar rather than in it. Good luck.

I pc’d for 25 minutes but my rubber plug wasn’t sitting right and it took a long time to get up to pressure. I do have a couple of drops of red fc in there but not much. Interesting. I’ll keep watching but the mycelium looks good and unaffected by whatever sediment or caramelization. I did not add sugar, does LME naturally have it?
