I have them sitting in front of my hood which is covered in a sheet to avoid dust.

No idea why it has these patterns.

I do this too often, I read too much and get confused, I have to ask a clarifying question.

I am reading a lot of agar recipes, I am trying to use what I have.

Is malt extract a necessary component or is it interchangeable with other carbohydrates? (Given that LME may be the best option, is it the only option)

Say, for instance, I have no malt extract to hand, can I swap in BRF instead? or do I need both LME and BRF if I want to make BRF agar?

I ran out of lme and used brf, it came out way cloudier but I may have measured a little off…nutes are nutes…i think some guys used to use dog food :lol:

No need for malt but go light on the brf , I'd go .5-.7 tops per 500 ml recipe

Hey! Do these look ready for grain?
These are Natalensis Clone > WA > T1..4

I'm wondering if all that fluffy layer on top of the myc is just tomentose or something else. The culture I got the clone from was really heavy on overlay, I don't know if that matters. Also, I feel like T3 actually looked more uniform and less fluffy than these, but unfortunately I missed the transfer window on those because I was away.



PS – that white 9 o'clock spot on the fourth plate is some sort of lens artifact or reflection, there's nothing like that there in reality.

Thanks San Pedro and cronicr, Got it Go lightly but BRF is useable.

San Pedro, I was looking at my dog's food the other day and wondering..."could that be used?" but I am way to new for experiments, but who knows maybe some day LOL

Dog food agar is definitely a thing Paul Stamets talks about in GGMM - I know most people shit on him here in the forums but I'm just sharing info.

pg90 10g agar 10g dry dog food 500mL "Dog food was first used as a component for agar by the late Dr. Steven Pollock."

sage, I am a complete noob so I am not shitting on anyone. I love more information and I am glad you shared this, and thanks for the list of recipes, I had found one in a search but not that one it looks much more up to date.

yeah np, lol I'm mostly just defending myself from any potential flack for mentioning him.

I'd recommend stick to something simple as long it works for you. Lots of folks I've seen use LME at ~2% with same for agar so that would be - 10g agar 10g LME 500mL water. If you're constantly changing your recipe / process it will be hard to determine the cause of any issues that pop up.

I screwed up last night and doubled the agar in the standard 500ml recipe.    I am thinking this batch is toast.  Looking for direction.

not optimal, but I would guess that you could reheat it and dilute then add the right amount of nutes to get it to target.

e.g. you were targeting 10g/10g/500mL - agar/lme/water (2% agar and lme)
but you did 20g/10g/500mL

So you reheat the agar so its liquid again and add in 10g lme and 500mL water then re-PC and you're set.

This is difficult for several reasons, namely, you're probably limited by your bottle size / number of plates you can pour. But you can always boil in a pot on the stove and pour that into two bottles if you have them  (I used a funnel when I used a pot but I typically just microwave in my media bottles now and no pour into HG plates).

I know reheating works for regular batches, folks talk about PCing their agar and leaving it, then tossing it in the microwave to heat it back up and pour assuming you kept it sterile the whole time. So maybe you save a second batch for later in your media bottle after you've poured your first set of plates?

Anyone please feel free to correct anything I've said here, I'm not speaking from experience. But I did work in a lab for a while and have read a lot here on the forums.

Also if you're microwaving agar - watch it the whole time or make sure its not going to boil with the time you set it to because if it boils over it will be a hot mess.
