Is there a difference between a spore syringe agar plate and a single mycelium covered rye grain seed and put it on agar.

I have cloned to agar in the past, and like you said i saw growth in 24 hours with pda.
But for some reason i cant get spores to go on agar. No problem when inoculated to grain.

T0 Yeti Plin Plate



I swabbed some of my first APEs, and the plates are good, maybe this a clone worthy pin to run alongside the MS and see the difference in yield/potency.



Another of the MS APE plates is showing a bunch of pseudocarps.



And here are some cool shots of a The Thing MS plate. This is a T1 of my mutant variety that sporulates. Now, it even though it is an MS plate, it seems to have passed down the mutant genetics as the dominating phenotype. Is this normal?





Just put some of those in tubs so I'm excited to see what happens! Been a bit slow recently.

Lil update on the fp+ scallop CS.. looking good.  Those off sections are sectoring, not scallops.  I’ll be letting it grow out some more and then taking a decent transfer to a new plate and letting that grow out all the way for confirmation it’s been cleaned


Interestingly, the transfer I took off the scallop plate before it started scalloping was put to a 4oz grain jar and I made a sandwich bag with it.  It seems to be doing fine.


I’ll do another sandwich bag of the culture after the CS to see if it does even better

A couple CS of a MiB clone looking awesome (I didn’t think at least one of them was dirty to start but still nice to make sure).  Excited to grow these out in the jcm

Thanks for the motivation @mackload and @kloakk! I got my shit together yesterday and did the damn thing! But here's what happened:

So last night I got everything together, cleaned out some HGs, and mixed up a small batch of agar. GF was helping, which really streamlined the operation. Triple-checked all my numbers - 350mL, 0.5% malt (1.75g), 1.8% agar (6.3g). Weighed everything precisely, mixed, heated, and pre-poured - using my new 20mL measuring spoon:thumbup:, 1 scoop each. Everything looking good, even amounts all around. Make stacks of 5 in the IP, place qt jar over each stack, add H2O and RUN IT! :dancer:

Then today I remembered I was supposed to stay up after the timer went off to set a 1 hour timer to time the cooling. So instead of doing that and wrapping up the stacks with towels to cool overnight, I just turned it off and went to bed. :facepalm3:

The carnage - Out of 15 plates, only 6 look anything like normal. They still have the same amount of agar that went in. But the rest... well, they were mostly full. :foreheadslap:  some had a layer of water, yes, but the rest had just absorbed that extra water and became a soft-set blob that took up all the space inside. I dumped them out of frustration before I could take any pics, but they looked silly and amateurish. And with only 6 potentially good ones, I'm hesitant to open one to check the firmness. I don't really have any reason to think those aren't sterile, so I'ma use em.

Pretty sure I added too much agua to the IP. One of the jars had filled with water 3/4 of the way up, probably when cooling or something. The bottom HGs on each stack were submerged about 3/8" when I opened it. I gotta figure out how I did these with lids on the jars, that way had the least condensation.

My first/best mushroom agar plate!  This is morchella deliciosa from outgrow. I plated a few other species, but this morel is lapping everyone else in terms of growth rate. 

What percent agar are y'all using?  I'm at 2% and was wishing I had a firmer gel when I was scraping samples for long-term storage.

^ Since you have the IP, I recommend trying the tek in my sig for pours. (the gnocchi water as well, I highly recommendm and it uses a waste product).

One run in the IP using 1000ml of agar mix yields me 80ish plates, using the sauce containers that I keep in the sleeve.

I used to do no preps, but the amount of work is insane. I can't believe I used to make agar for just 10 plates at a time.

edit: I've had problems with everything else, but not much with agar. 20g agar, 20 malt per 1000ml water for 2% MEA, mix on stove, pour into media bottle, IP 15 mins, let cool til comfortable to handle, pour in SAB. don't worry about condensation (it mostly clears, flip plates for wetter ones, doesn't matter terribly if it doesn't)

Oh man, that is not what one needs when it's already hard to get going in the first place :wow: I never done no pour, but I've had my share of trouble with spilling, pouring to hot or to cold. Yesterday when I had a SAB session the parafilm ript and I bumped my arm in the armhole so hard I could feel the draft of air on my face :mad2:

Did what I was supposed to anyways and had a few beers to chill, but yeah I just wanted to yeet all the plates and jars and walk away. But I'll be back pouring more plates soon cuz I know I have new genetics coming in :rockon:

Some days just don't sit right and that's okay, fuck that day, I'll try again tomorrow. Just wanted to say I feel you man :muppet:

I highly recommend getting sauce containers. I have not had contamination issues, they come clean enough. They seal, don't require tape, stack and are recyclable. I grab a sleeve of 100 for like 2 bucks at a restaurant supply store.



Plus the hot agar creates some japanese wave paintings patterns

Yeah would like to try, but in my country I have to order them online and with shipping that will set me back $20 which is the same as 100 petri plates..

And I was wondering, how do you pour them? Like, do you line them up or keep them in stack?

So I keep them stacked inside the SAB, with the cups bottom up, lids top up. I sacrifice the ones at the very bottom because they touch the surface.

So I quickly take a cup, flip it over, pour agar, then put the lid, bring it to the side, rinse and repeat.

I have had 2 specific plates contaminated with Cobweb + Ochra mycelium from swabs. It was the first time I saw it, but I knew what it was right away. It appeared overnight after inoculation: the germination speed was terrifying.

In just a few more days, it took over the entire plate.

Whats interesting though is that the Ochra spores also germinated in both plates and started to grow nicely in the center, not caring about the already densely present cobweb! :shocked:

Here they are at early and late stages.
