Before cabin and trench sequestering caught on sandman420 used to talk about a similar idea on discord but just putting the transfer on the wall of a no pour or deeper agar vessel and it would do the same thing the myc would grow down off the wall onto the agar and leave whatever else behind cuz it couldn't grow down the wall

Exactly there's always nutes left behind but bacteria doesn't travel well on a dry surface but mycelium doesn't mind seeing what's around the corner

How I understand cabin sequestering is that bacteria won’t grow through.  It grows on the surface but doesn’t burrow.  The same reason cold pours and trap doors and sandwiches work.  Unless I’m completely misunderstanding the intended use of them.  I don’t think I am though. That’s why I asked if the myc could be pushing the bacteria through with it.

Technically the transfer inside the cabin is upside down but in my mind, when you make the cuts for the transfer, you’re pushing fragments of whatever was on the surface down to the bottom anyway.  Unless the transfer in the cabin is supposed to be shorter than the thickness of the cabin plate so it doesn’t touch the roof so it grows along the plate bottom and then up the insides of the cabin.  That doesn’t make sense to me tho.. why even have a roof at all then?

If it’s because the meeting of the cover slip and the rest of the plate is supposed to stop bacteria, why wouldn’t the roof itself stop it unless it’s being pushed through.

Well, the way I understanding it, the myc is supposed to grow between the cabin and the roof, not through the roof. Furter more, glass is not as attractive as agar.

Have you read MycCycle's journal?

https://www.shroomery.org/forums/showflat.php/Number/29049976

I'm just saying, scalloping is one of two things, backy or monokaryon, what do you think your plate is of the two?

I have not seen that.  Appreciate the link.  I was going off of supa’s thread where agar was being used as the roof.  First time hearing of the actual cover slip

Agreed, so if it is indeed bacteria, then cabin sequestering is the best way to eliminate it right?

Or did I just see someone do that? I still have no idea what it’s for.
Seems like a lot of effort to replace regular careful culture cleaning 🤷‍♂️

Ive been trying to figure out how this isn't just trenching with extra steps. Im not saying that's all it is, I'm just saying that I dont get it. Ive seen the diagram and whatnot, but I dont understand this "myc growing THROUGH the agar" thing.

Is the transfer supposed to stretch to the walls of the cabin and then grow up and around the roof? That's just trenching with parkour, no?

I also saw on a sequestering write up that the glass slip was a crucial component, and I told myself I'm done buying things that I need for a single application. Wooden pickles aren't selling so well since Christmas is over and I'm fuggin broke.

Thats the way I understand it as well, but only from something I read on this forum once.  It's the same principal as a trap door which is where I read about it.

So assuming this is true, then its not bacterial I would expect.  I guess the real test is, how does the next transfer grow?

Just a month or so ago everyone talked a lot about using BRF pucks to clean bacteria off a sample since the bacteria cannot travel across brf.

No problem, I just want to do my best to get my FP+ clean :awesomenod:

I guess the cabin sequestering came to this community through supa and he is getting it from Paul Grzybowski, now Paul choose to use agar as a roof, but the original technique never mentioned agar, only coverslip.

I do not know if it will make any difference, but what I do know is that I could get high and trip balls to see the meaning of life and ego death any time.. Been and done, I'm not in this for the high, that is just a bonus.. But being part of this community and actually making a difference, well shit, that would make my fuckings day... So, as soon as my T6 FP+ plates grow a little more, I am excited to try the tek and see what it can do..

It’s also pretty early.  This may have been premature.. it just looks like it’s going to scallop but I’ll revisit once it’s grown out more.  But I will be trying just the sequestering too.  I’m not sure I get the glass cover slip though… there’s nutrient agar underneath it.  Why would just a piece of glass stop bacteria, especially if the bacteria is riding along with the myc?  Idk, maybe there’s a mechanism going on I’m just not getting atm.  I’m about done thinking about it for now lol

Yoooo, the both of you, have you seen the seminar? Both your answer is talked about there..

@Thurman, the roof will stop the bacteria because the mycelium will grow between the roof and agar.I just bought 200 slides(that I will cut in half) for 3 dollars. You can afford it.

If you check out Paul's seminar then you will see that antibiotic agar isn't helping anything, it's just producing more antibiotic resistant bacteria..

Here you go boys:

You're telling me, man I've been drinking for 10 hours and soon I have to get rid of all the fucking snow outside so I can celebrate my daughter's 7th birthday. Beer is what is giving me the courage to even discuss this with all y'all OG's

But I really want to understanding this, and be better, I don't care how or by who, but be better at cultivation.

Same.  I think it can be a great tool at fighting bac.  It was great for my semp clone

That seminar is what got me excited about it

Thanks for saying that, to me you are light years beyond me, and FP+ is my nr.1 goal to clean, and knowing we have the same goal is really good. I know, I am John Snow and I know nothing, but I really want to know:lol:
