Not necessary.
But are there benefits, probably.

Tnx. What would those be, besides what sage_bd said below?

I don't think the wood does anything for cubes, but it does seem like a convenience thing. It seems like it would be easier to work with since you can pull the stick out with sterile tweezers then grab a transfer.

I have slants with popsicle sticks in them but the only thing I have on them right now is wild turkey tail which I think wood is probably beneficial for. Make sure you hydrate/sanitize the sticks in boiling water before use.

Tnx mate. I don't know if I'll be using sticks though. Unless I hear a good reason to.

And when we're on the topic of slant's, has anybody ever made small, micro slants using these? I really don't have much space in my fridge ... I'm wondering really, what are the benefits of the bigger centrifuge tubes vs agar plates anyway, except that centrifuge tubes close them self's up better and thus maybe don't let as much moisture through (all my agar plates in the fridge end up having water in them, in spite of having them locked up in sealed Tupperware).

I've got a whole bag of those and the little stand thing they fit in
Still never used a single one

I would only use the Pyrexx. I have heard of bottles exploding in people's hands due to rapid temperature changes. Beyond trying to give people good growing advice, my number one priority has to be safety.

I know you don't need this explained to you but just putting this out there.

Rapid temperature changes with glass should be avoided even if you do have pyrex. I boiled out all of my solvent in one of my labs during college and tried to add some water back to the blazing hot pyrex beaker and it shattered (obviously).

Glass isn't designed to handle rapid temp swings. It's a poor thermal conductor and isn't a malleable material (unlike metals which still warp from rapid temp swings).

My guess is that these would be too small to serve the purpose that slants serve.

At least in my understanding (which is limited) the purpose of slants is to preserve myc for an extended period of time and having a 15mL tube slant with approximately half that filled with agar and slanted, it gives a large pocket of nutrition for the myc to feed off slowly over the next 6 months to year before folks recommend performing transfers to maintain viability.

So by using those tiny centrifuge tubes, you don't provide many nutrients to the myc for preserving it over a long period of time.

But anyone feel free to correct me here, just speaking from my limited understanding.

I initially bought them thinking I would try some distilled H2O storage, or potentially storing dried grain

Might be able to use eppendorfs for something like Cryovial Tek

I mean, mycelium in the fridge doesn't use up much nutrients as it's growth is more or less stalled, so, idk how much nutrients it really needs?

Dried colonized grains you mean?

Ah, there are sooo many ways to go about anything in mycology ... If mycelium can be stored in distilled water without any nutrients, outside of a fridge, then why the heck would you want to bother with slant (I understand slants have benefits) ... Appreciate the link!

I think there are better ways (than slants) to store long term, here are a few links I'm aware of.

Easy LI for Preservation and Propagation
Cryogenic storage of P. cubensis mycelium, or can I freeze my cultures?

I’ve used these to send wedges of agar with culture, and have received these from others. I have a bunch that are almost a year old with usable cultures still inside. I’ve stopped using slants. There are other easier ways to store cultures.

Welll anythings possible but pyrex is designed to handle some major temp
swings.  think about glass-top stoves.

im not getting anywhere near 40 plates at 1500ml.  i could count them later when i get home but i think thin plates suck.  they dry out too fast and they tend to stick to the walls of jars bad.  i like thicc plate.

Seeing all these great looking plates in this thread makes me question my capability and methods - I'm never really 100% satisfied with every plate or every poor and I wonder if this is similar for everyone.  So far I've only worked in a SAB, methods I've tried - rated from memory:
- Started with disposable plates and pour method - great for clarity, good for condensation, fair for contam (bad for environment!)
- Tried reusable glass petris/pour method - great for clarity, fair for condensation and contam
- Tried PP5 90 mm  petris/pour method - good to fair (opaqueness with reuse) for clarity, good for condensation, fair for contam
- Moved to no pour with the glass/PP5 90 mm - fair (opaqueness with resue) for clarity, fair for condensation, good for contam; also occasionally had trouble with agar (re)solidification
- Latest choice is using the 'Primo Plates', no pour tek with 60 mm clear Viabooth petris - great for clarity, fair for condensation, great for contam with nice benefit of bullet proof storage prior to use.

I recognize that some of the contam issues may be my SAB technique (big, clumsy hands:rolleyes:).  Some questions: Are my expectations too high? For those that are satisfied with their method of choice - are you still pouring 15 to get 10 good plates as an example?  Also, if you started on your journey in SAB with no-pour then moved to FFU did you go back to pour method with improvement on plate condition/yield?

I'd gladly shell out for an FFU in exchange for 100% good plate pours.

Maybe it’s just me but I didn’t quite get if your main problem is getting contamination even with plates you haven’t even used yet, or problems after inoculation, transfers? This probably influences the discussion quite a bit.

Edit: Just as a data point I just started with agar. Have poured 60 plates in a SAB (mostly because I’m quite paranoid about what I’m transferring). And I have had two unused plates that had something on them, and I’m not sure if one bacterial weirdness wasn’t just introduced during a transfer.

Agreed and the only time I'll ever pour thin plates is 60 ml dishes I plan to use for lc ( I get less condensation with thin plates with the 60 clavables)

They dont even need to be in the fridge.
I've had better luck with older wedges in these ,Than I've had with newer cultures sent in Dh20.

I've gotten a few plates with what is clearly edge contamination when using the pour methods, I haven't been sealing after pour/before use but I have stacked and covered them in the SAB. I've also gotten the occasional random splotch which I attribute to my SAB/hand technique when pouring(all before use).  I didn't consider these a huge problem - just another factor that lowered my yield of good, usable plates from a pour session or technique.  With the Primo Plates tek the contam issues have all gone away but condensation is not satisfactory (for me). Were all 60 of your SAB pour plates usable?
