Yeah, Iā€™d say the benefit is speed since you have more tissue on a wedge and ease of identification. Easier to see where the good sectors are šŸ‘Œ

First time growing and first time playing with agar.

Went spore to agar, then transfered to agar.
Used potato starch and dextrose recipe.

Did 14 plates, but agar turned too cold in the end and they became lumpy and not clear.

So my question is, which look fine to take to grain and when?

Or do I need to clean it up with more transfers?

They all look off, do you have the original germ plates still.

I was planning to make some agar and pour some plates last night and this discussion got me thinking about my extract choices I had on hand so I paused to get more info from the experts here. 

Trying to solve slightly erratic differences between agar batches that I think nutrient level is the main cause.  I have 2 different extracts in inventory: a bag of 'LME' that I purchased at a bulk store with no other info other than the tag I wrote when purchased and a jar of 'Dry Malt (Diastatic) Powder' that was originally purchased for bread baking.  This does have nutrition facts label with following info:

Ingredients: Malted Barley Flour, Dextrose, Flour (contains Wheat Ingredients)
Serving Size: 14g
Calories: 52
Fat: 0.36g (2.6% of serving size)
Total Carb: 9.5g (67.9%)
Fiber: 0.6g (4.3%)
Protein: 1.1g (7.9%)
Sodium: 1.1mg (0%)
Sugar: 4.1g (29.3%)

Note: I calculated the % of serving size - this is not the daily value % required to be listed. I've never been real happy with the bulk store stuff from a particles/filtering/clarity standpoint and the diastatic stuff is much finer so I was considering using it again but the sugar seems a bit high so here are my questions:
- Does your 'go to' extract have nutritional label on it; if so, how does it compare to this?
- Doe anybody use these values to calculate nutrient levels actually being added or is this too much minutia?

Yes, i have the plates i did prints on.

Some of these posted here the grey spots are from tiny agar pieces from the transfer.

Can you elaborate on how you notice it is off?

The agar seems fine, no bad growth from the pour itself. Had them staying 1 week at first when doing from spores, and no growth. So seems like i have the agar pour technique down.

But diastatic malt powder contains (usually mostly) wheat flour. Not something I would want to put in my agar. Flour is mostly unconverted starch and gluten, neither of which is what our mycelium prefers to eat. The wheat/barley malt product Carnonos mentioned is the result of mashing wheat and barley, then spray drying the resulting liquid. The mashing process converts the starches to sugars we use as fungi food.

To answer your question, no. My DME doesnā€™t have nutritional info on it, but it can be researched easily enough. No I donā€™t calculate anything other than the percent of DME in my agar as it related to the volume of water used. I use dry malt extract intended for beer brewing, the lighter (color) the better. Nutritionally itā€™s all about the same. Beware not everything labeled ā€œmalt powderā€ is barley malt extract.

It's the growth that seems off, not anything about the agar (pour).

It's not uniform some kinda remind me of monokaryons, which can also resemble some molds.  Not that I'm say these are, but it is possible they may not be clean.

And I wouldn't take those to grains.

So more transfers, from leading edges.
Take Small cuts from the most uniform/ organized mycelium


It may be a good idea to start more germ plates

Y'all helped me figure out how to kick the crayon eating and work a camera a bit, so here goes!

This was gifted to me as BAPE (rust revert). I haven't fruited it yet bc the myc has always looked like this. Im pretty confident I was told months ago that this is normal, but I wanted to verify that if I could.

The agar is of inconsistent thickness, with the top being the thicker part. šŸ˜¬

Yes, but not necessarily just technique.  It could be the spores your working with too.

Once you get germination, post some pics before transfers and we'll  help you narrow down the better sites to cut.

Those look like they are off to a really nice start. Rice can be a tricky grain to work with, but it looks like you nailed it.

Hello All,

I have a transfer question.


I took a tiny GT 4th TF at around 12am on 9/1 and it's showing some nice vigorous strands.



This is the plate I TF'd from for reference:




Qu: Is it too early to grab those little bits for new plates or is it worth growing them out a little more first? They have a day to develop with heat and light until I'll potentially dive back in.

I understand more about sectoring since reading this thread and so I think it's important to say I'm not a noob hunting for a monoculture, just a nice, strong, evenly balanced plates.
