Ok, that makes sense, but then what is the answer if someone asks which classic cube strain is most potent? Bc I thought the answer was still "a cube is a cube" and it had more to do with specific genetics and not the variety itself. Again, I'm not taking a position here - I'm just trying to understand.


We will never know what's the most potent , what's the most potent in your own collection?
I've grow tidal wave that ripped my head off and tidal wave that was kind of meh.
If we tested 100 people's GT's  would every one of them come in at .8 percent?



Thats exactly what I was told many years ago - whatever variety name happens to be attached to the spores in your hand is irrelevant- all the fancy names are essentially marketing BS and they're all "just cubes" and in order to consistently get high potency, you'll need to select for it like any other trait.

With that said, I understand now that "a cube is a cube" was more about the actual cultivation techniques.

Regardless, I apologize for adding misinformation to the thread.

If you're using the kit but not the Colorimeter, is there a certain light color temp you should use to view the results (with white paper behind, curved, as in their video)?

I think different light temps would give a visually different reading with the eye.

This is why you isolate clones. I have cultures that reliably test at the top or above set of contest results. The new availability of test kits helps sort this out earlier.

Which non-PE Cube varieties have you isolated and what are they testing at?

Are you using the PsiloQ test?

I read a compelling case that Nektar wrote about how you get better grows by getting cultures with good symbiosis of culture (my words), I think is what she meant by the sectoring thing? getting 2-3 sectors or some such.  I didn't follow it all.

If you mean the thing of "finding single sectors of agar to produce a monoculture" that's old tek, wrong, and b.s.

We had a sub-thread on that topic here a few weeks back. Some TCs used to even spread it, but it's voodoo junk science.

Some people still spread that, so it's still around, but it's bunk. Like "separate fruiting conditions" and putting coffee grounds into a grow. All ancient stuff that's no longer best practice.

Yeah, you were saying that having 2-3 sectors?  I think that meant, it is now recommended to have more than one spore genetics getting along with each other on agar before going to grain?  I haven't deep dived it yet, since I'm still waiting equipment to arrive before doing agar myself.

You shouldn't think about "sectors" (or related "monoculture") when transferring mycelium to a new plate. That's old, wrong info. (you might be thinking of something I wrote debunking the idea of "monoculture" or "sectors")

You should only be looking for clean robust mycelium, and transfer a small piece of it.

When you get your supplies, put spores to agar, and have a germination plate about 1/2 to 2/3 grown out, post a good close pic of it (or of a few plates) on the Agar thread and ask where to transfer from, someone will recommend where on it transfer from on the plate. (Usually as clock position, like "Outer edge at 9 oclock") 

After you do that a few times, you'll learn to recognize it.

I have cultures of PES amazonian that consistently test above 2%.  I have a KSSS culture that runs about 1.8%.  I use the Miraculix tests.

edit:
i suppose they dont look horrible now that i got em in the light but, no where near the beauty u got goin on. my plates tend to look questionable until they colonize 1/3-1/2 the plate. anyone else experience this? i probably should go back to using powder lme instead of this syrup i accidentally bought. especially since im using the same amount i would if i had powder hah
