Honestly would be fine to just copy and paste his, credit him, remove that part, and mention a wrong, out of date, part was removed.

Or mention anywhere it's linked that a lot of people will see it that one tiny part is wrong and fine to ignore. I've seen about 5 people saying they're looking for "monoculture" "sectors" lately, at least one of them found it through you linking Stro, and I bet a few of the rest did.

There are also probably people quietly looking for sectors after reading Stro right now without mentioning it too.

Thurman said he wasted a lot of time on it before he happened to mention it and was corrected.

If Stro were here I'd talk to him about updating it but he hasn't posted here since 2018.

A lot of people do that part even though it's been found to be wrong.

Ok good be on track then. Skipping the strain isolation stuff 👍

The recipe is just plain old light malt extract with I pinch of nutritional yiest. Nothing fancy. 20g agar and 20g LME per 1 liter of distilled water.

I took three more tissue samples from the same tub after the initial one with the rhizomorphic, but they aren’t branching out the same way. I got the rhizomorphic growth after cutting out the whole tissue sample from a multi sample dish and transferred the square to a new dish. It grew down along the edges and stalled before shooting off in all directions.

Plate nr 3 is behaving differently. Looks like it is shooting upwards vertically. This is day 7 from cloning. Any comments here?

I will probably cut squares off the other three plates and se if they also shoot rhizo if transferred to their own plates.

Hello All,


I am looking at this plate (stored in the fridge, hence the date to growth ratio)
and I see several things that look like potential contams.



The gap at 12 oclock looks like a void



The area highlighted with a white rectangle looks slightly green/blue which I'm guessing is mould of some kind.

Am I right?

Nothing to worry about there, IMO. The section at 12 o'clock just is just less rhizomorphic, and the discoloration is most likely bruising (from drying out or a temp swing - which also can dry out an agar plate).

Just make a new plate from growth on that plate that looks healthy and organized, which is most of it, especially in the lower half.

I also concur that chasing sectors and monocultures is an outdated and unrealistic objective.

Things should be updated to the new state-of-the-art, which is compulsively cloning plins until you're drowning in a bunch of closely related cultures that you can't bear to part with.

Ok, maybe that's just me...

Why do you prefer plins vs actual fruit from a dense pin set in tubs?

The main reason is that I'm pulling from a plate that hasn't been exposed to open air, so it's very easy to just pluck a whole pin and drop it onto a new plate, and then I've got relatively clean growth to transfer within a few days.

It also keeps cloning work completely separate from tub work, I'm busy picking and processing my tubs and don't want to think about choosing particular fruits to clone from a sea of options.

On a related note, I can do cloning and culture development before I've even gone to grain. If I want to test stuff, I can just crank out a bunch of clone lines and then put them to grain simultaneously, and see which ones I like. (Only problem is that I usually end up liking most of thm)

But practically speaking, I don't see it as terribly different from cloning clusters out of a tub. I often get clusters of plins, and my philosophy is that genetics which will pin readily in a petri dish will go absolutely apeshit in proper conditions. That has pretty much been borne out, in my personal experience.

But also, big picture, we're looking for good genetics and clean growth. A plin constitutes all-new material somewhat separate from the rest of the plate, and genetics that will obviously fruit. Checking a lot of boxes in one transfer.

Interesting, thanks. I'm finding a lot of my transfers from areas of strong, visually similar rhizome growth grow out to look similar to your example.

Do you put plates like your example to grain or aim to get myc that is similar looking all the way around first? I imagine both have their advantages.

I’ve been trying to work with a 4 year old lute print that I have and cannot get to germ. Even forced hydration to agar is showing nothing. I just hold out hope that someday I’ll pick up that plate and see activity. 

I’ve tried, warm pour, forced hydration and brf puck to get these started. Nothing yet. Any advice?

I send them to grain. As long as all the growth looks healthy I'm not worried too much about the sectoring. I'm sure there are differing opinions here. :smile:
