Cutting bacteria out of agar plates? [Re: Tormato]

Noticed a few of my freshly poured agar plates had a spec of bacteria growing on the rim where the agar meets the glass.

Anyone ever just cut a significant portion of the agar dish out around the bacteria and continued to use the plate successfully?

Yes and no. I’ve cut contam out of an already colonizing plate, but that contam was the introduced when I was making transfers, not while pouring. I wouldn’t trust a plate that contams before use, personally. There might be more brewing than what you see. I call those plates loss, tbh.

i've been seeing this stuff from time to time. last batch of plates i made seem to have this in all of em. I don't see it break the surface, so i always thought it was something like separation or the like. my old lady says she thinks it is an organism. if it is, my pc might be fucked or something. i run these for 90minutes.
anyone have any insight?

Looks like something has settled.  Is that from scratch?  Is it in both the first and last plates to be poured or just the last?

i'm not sure what you are talking about. i haven't touched the subject line. i'm looking at it now. it says Re: re: AGAR ENVY! (Anything and all things agar!)

these are no-pour from scratch. water potato flake and telephone agar with some food coloring. i added a bit of nutritional yeast cause i'm using these for lions mane.

edit: oh i see now that everyone has "cutting bacteria out of plates" as their subject. i don't have the option to change it to anything else. aside from typing something new in

Looks like some mycelial growth in these areas or is that just the pic? 


something could be precipitating out and settling or perhaps that’s the bottom of the pan/last cup to be filled and got some sediment?

Based on the pics that’s a blank plate.  If there’s something growing there, it’s likely contam.

the stuff you circled is soap/hard water stain on the outside of the plate.
i haven't applied anything to this plate yet.
if it were a contaminant, would it stay under the agar without breaking the surface?
when i shine a light across the surface i can't detect any inconsistencies. looks very smooth.

i am leaning toward something precipitating out as that seems the simplest explanation. otherwise my pc isn't getting up to temp after a 15min purge. thing isn't that old. i've had it for maybe three years.

my potato flakes are old. several years old. old corn syrup too. but i don't add enough to make something this big. and it is present in multiple plates

Switch up to LME.  It’s all you need and super simple. 2% nute, 2% agar.

Liquid culture would use .2% nutes.  Standard agar media use 2% nutes and 2% agar of the total volume; water is 1g/ml so 200ml of medium would need 4g each nutes and agar….LME (light malt extract) is a nute and can be purchased dry online cheaply.
