I have screw tops that I didnt put vents in, loosened it quite a bit, and they still got crushed ;_;, i put vents in screw tops now

So far, I've been lucky with mine, except the one I didn't loosen enough. That one imploded nicely I leave them fairly loose now.  Do you wrap your cups in foil when you pc them? I did a few times and still got a fair amount of condensation. Might just stack them in the PC as they are with a sheet of foil in-between the layers of cups and see how that works.

No hole needed. Be careful opening the container to do transfers. The lids can splatter drops of water everywhere. And if you flex the lid to far they can crack. Not sure why, but I went from a 50/50 contam rate to 90% clean when I would open them upside-down too. Oh and careful if you have moisture and it runs around the lip of the cup don't let it back down to the agar. I used a paper towel in my SAB to soak it up. And watch the inside lid lip for moisture too.



I was wondering if I needed them. I haven’t done a no pour yet but love the idea.

I do foil them.
I usually take them out of the PC hot, remove the foil and stack them ontop of each other as they cool. It lets the agar reabsorb the moisture as it condenses. I put a cup of boiling water on the one of top.

Sounds good, going to do it that way tomorrow. I also saw a thread where they put a baking pan on top of the cups and poured hot water into the pan to keep them warm as they solidify.

Not my normal cloning method. I had extra plates laying around and threw some old mushrooms on them. A lot of strains in there.

Many people would let that grow out half again as big, but most people don't let it grow all the way to the edge to transfer.

Point of transferring at this point is to get clean mycelium, and to get good looking, well growing mycelium. You can do both at that point you show there, but some people wait. I don't see the need to. You've got organized rhizo-y looking mycelium on the edge, and it looks clean. Waitging longer lets any hidden contam grow out.

I let it grow out more on the T2 or whatever I'm sending to grain, because I want more mycelium to send to grain. I usually do 7 quart jars at once, because that's what fits in my PC.

But if you're looking for transfers, I don't see the reason to wait.

I would do it now, but nothing will be harmed by letting it grow 2 or 3 more days (depending how fast it's growing.) 

Anyone who replies contradicting all this, I believe is showing
a personal preference, not science, and should "show their work" and say why they let it grow out more before a transfer.

Growing all the way to the edge can lead to contam too, if you're plate isn't entirely sealed.




This is very useful. Thank you. So my plan with this Mak is to get a couple transfers out of it and then slurry the rest and send to grain as a MS run.

This point would be sufficient to get that done?

My Saturday morning project. 2% LME, 1.8% agar, no peptone this time. Telephone brand seems to set rather stiff so we'll see how this turns out. That and I have a few swabs to germinate so I wanted a slightly wetter matrix. I don't have a pic, but the condensation was not too bad this time. I placed a sheet pan on top of the cups after PCing and poured hot water in it. This seems to have helped.

It seems to have reduced the condensation as compared to my last batch. Next time I'll use hotter water and maybe a deeper pan to reduce it even more. As is everything in this hobby, it's a work in progress

For no-pours, I do batches of 10 at a time. To "fix" condensation what I've been doing is boiling water once the cycle is close to ending. I take the no pours out of the PC while still hot and I take other no pour containers that are unused, put one on top of each of the ones with agar and pour boiling water on top.

It works super well and relatively easy for small amounts of no pour containers. Easier to handle than the pan full of water 😅
