What is your process for creating GSW agar?

To the best of my knowledge, you need to cook grain water agar for longer than LMEA.
You'll want to use grain soak water within ~24 hours.

Munchauzen, I think, stores his GSW as ice cubes.

I made the GSW agar plates from water that was boiled with oats (with added gypsum) for 30 minutes. The water sat at room temp for about 2-3 hours before I reheated it and added the agar. Sterilized at 15 PSI for 30 mins

Yeah I’m really having trouble understanding if this is contam or not. Doesn’t look like any bacteria or mold that I’ve seen before

Perhaps I should have made my own post about this, if this wasn’t appropriate for this thread then please accept my apology everybody

For the parafilm users, what are you doing to cut it? Are the dispensers any good? They seem super expensive and have no reviews, or the cheap ones have terrible reviews. I love parafilm, but I’d love it more if it were easier to cut a 1”x4” piece off the roll.

This is the best way to cut your parafilm in order to avoid microtears. Other lengths/widths are highly prone to microtears in your film and thus a vector for contamination.
4 squares long and half a square wide allows you to wrap around the plate 3/4 times.

And this paper cutter is so convenient because I can cut hundreds of pieces of parafilm in such a short amount of time just by stacking sheets and cutting through multiple at once.

A question about agar is 100% appropriate to this thread. Even more so than half of the asides we all go on.

I don't have an answer for you, but I'm wondering if it could be metabolites. I usually see those on grain or tubs, and they can be caused by any stress to the growth, bacteria, mold, too much water, too little water, too much air, too little air,  etc. Maybe too little nutrients.

I haven't seen metabolites on agar. But there's always something new with so many people doing this hobby.

How long did you PC the agar mix? If more than about 30 min, it could be caramelization. Not dangerous, but cut down to about 25 min next time (unless you're way above sea level).

There's enough air in even a sealed plate to be enough for full growth, according to Bod, so maybe something else. Other than the yellow it looks good. I'm stumped.

Personally, I'd transfer away from several of them to new plates, from the edge where there is no yellow. If I were felling particular bold that day, I might even put some to grain, from the edge where it's not yellow.

When you open it, if there's any smell other than mushrooms, don't put it on grain, just transfer it.

I like your user name. Made me laugh.

I’m wondering if it’s metabolites from it changing food source, different genes and enzymes need to be used to break down different food types.

Sometimes people change up agar food mix, supposedly it invigorates the little mycelium from getting tired, like when using a clone of a clone of a clone etc.

This isn't the best thing I've seen on it but:

I use the same thing all the time, and almost always have rhizo, and often vigorous rhizo:

I'm all in on parafilm. I just use scissors, cut two squares per plate, and have all the precut pieces inside the SAB.

I fold them in half long ways. I do hate getting it to separate from the paper though.

I also ordered the 500 count box from southern labware. Mine came in sleeves of 20 though, which is not convenient for me, but I'm making work.

Honestly I tried both ways forever. Would always end up with micro tears in my plates.

A hugely successful commercial grower had the same issue as me (it's very common apparently) and recommended the cut in my pic. Completely solved the issue and I prefer adding to and removing it from plates this way.

Umm… maybe that was my problem and why I was getting so many holes that develop while the plates were sitting on the shelf. I got the single square roll and was just using half a square to wrap a plate. For the last few plates I’ve use a full square and folding it in half, then stretching that around the plate.

I recommend 4" parafilm and cutting 1/2" strips across with a pair of scisors tip: stop the cut a few mm before cutting through when cutting multiple strips to make it easy for multiple strips. Then you have a fan of strips you can snap one off of.

The parafilm comes off the paper much easier with 4" parafilm and 4" is enough to wrap anything.

I agree with both of you that was my first thought initially. I had about 20 cultures with 3 species of psilocybe on LME and when I moved them to the new media 100% produced these colors. This is now 2-3 transfers from GSW to GSW still acting like this.

I’ve heard that mushrooms do have to adjust there biological process when they encounter new kinds of food. If that is what’s going on I’d expect somebody to have encountered this scenario before though!

Here’s a picture of a dish from the same batch that was never inoculated with anything, stored at room temp for about 2 weeks. I’ve heard about carmelization but never seen it so I’m not sure what it looks like. This is unfiltered grain water so there is some sediment. I don’t see any contamination growing.

I have wrapped many dishes with 4 x 1/2 square lengths of parafilm and agree I think it may tear less than 1 x 2 folded in half but IME does still tear occasionally. I actually went back to 1 x 2 as my standard because of how annoying it was prepping so many pieces of 4 x 1/2 individually with scissors. I just purchased one of those paper cutters online, I’m going to give it a try!

prestretch before wrapping to prevent tearing. Never tears with a good prestretch.

Difficulty of wrapping plates isn't the issue nor is it even the subject at hand.

It's the simple fact that parafilm can be prone to tears. Nothing to do with the way a plate is actually wrapped.

I'm not talking tearing during wrapping.

It happens during long term storage, it can be weeks or even months before you notice micro tears.

I've stopped keeping plates around that long

And if you are keeping plates around for a long time and actually working with them then they get opened and re wrapped during that process.

And if youre not actually working with the plate but instead just holding on to it then probably wise to change the out the parafilm every so often

I'm keeping nutrients down to 1% for a while now and don't see any mentionable difference in performance.
Germinating spores is working fine with this percentage as well. Will go and check out 0.5% soon.
