You can use yeast without sediment. I use Bragg's but I'll add it to my water and bring it to a boil while stirring and filter it through a coffee filter. Throw it back in the pot, add my agar, throw it in my media bottle and PC.

LostKeys, I use the peptone I addition to yeast, not in place of. Peptone, LME, and yeast is a great combo. Also grain water, peptone and yeast.

I never liked using the scalpel. Not that the scalpel doesn't work. It always worked fine. Just on a micro scale I always felt any transfer I did never isolated just the rizzo strands I wanted. when they did make the long journey from plate to spawn, or whatever.

Found Josex's Poke tek. Josex uses a sterile syringe with sterile water to do his transfers. It was a game changer for isolating strands for me. no muss no fuss. Poke and drop. (Literally one drop.) And well isolated cultures began to appear before my rookie eye's.

I do all my clones with a syringe now too, haven't lost one to tams yet. For Lc's Squirt a few ml's of water (in the colonized agar dish) scrape (agar) and suck all the myco goodness back into the syringe. flame sterilize, Then noc up your LC.

PC Foil wrapped syringes tip up when you run your Agar and LC solutions. (same time needed) There is so much less risk of tams. Thought I would share.

I've dropped wedges of prime culture through my grating making it un-retrievable, then cried. No more tears for me. Good luck.

If you whisk things around while filtering it'll be done in less than a minute.

I think I'm seeing things. Does anyone else think placing a hot mug on a plate "flatten" wispy growth on a germ plate or early transfers?

Just took some T1s of Ghost and Jack Frost tonight which I'm excited for..

A Ghost revert and PE clone looking nice on plates..

How's this look for Lion's Mane? Not sure if it's a clean culture or straight up contam mold. Came from a dried out plate that someone sent me and it sat in my fridge for a few months.
Sorry I know the LM pic sucks. I'm realizing glare is a real problem with plastic plates. Probably will build some half-assed light box soon for good record-keeping

originally ordered Celltreat grip ring direct, but it is cheaper from Amazon and the shipping is free instead of $50.

About plates, ffs, i poured like 80 couple a weeks ago. Got hit with the headaches that sometimes plague me and couldn't work. They were decondensing in the flow. Went to do transfers last night and they had dried out and gone from 5/16^ths-inch thick to like 1 mm. :facepalm3:

They are tough to get from Amazon right now. It's where I bought mine previously but many sizes are out of stock right now or have shipping times of 30-60 days.
