Had the worst luck doing it that way, just the sloppiest wettest plates.

Jumping on the “keeping plates almost vertical to combat bacteria” train.  Digging it so far.  Should be able to easily snag a top transfer and run it through a cabin.  MiB clone


Also think I snagged a good transfer with this swipe through some aerial myc on the top of a botched cabin (more of an experimental trenching/cover slip than a cabin.  Tried using a whole stipe under a cover slip).  So far, so good.  Here’s the cabin..


And the transfer.. I just drug my scalpel tip very carefully through the aerial myc without touching the agar below it to get some myc on the scalpel.. helped using a number 12 blade (the tip hooks downward)


Even if bacteria does show up (I think I’m in the clear.. I would’ve seen it by now based on how fast the bacteria showed up on the cabin plate), they’ll be lots of lead time for the myc to run clean to get enough for a new cabin.

Me again with a query. I have two cultures with a really light growth. Not sure why I let them to grow out that much, I guess I was hoping that these are still usable.




I’m imagining that it could just be because of the nutrition of plates. Got like a full gradient of colors when pouring, because I didn’t bother to swirl the media a bit after PC-ing.

Or it’s just a shitty culture. Or maybe there’s some sort of hitchiker there, maybe it’s just mold? Though nothing seems especially off to me.

I should probably take a transfer and see how it goes on another plate and not use these, right?

Hello!
I'm a noob with some questions that I wasn't able to find my answers to (or found conflicting answers). I recently got a swab of P.Nats (or P.Ochra) that I swabbed on some plates. Now, the plates are growing, especially this one, and I am wondering if it is supposed to look like that? Also I've heard one should start with doing transfers before inoculating a jar, but it is unclear to me how many I should make before going to jars. I was thinking on eventually doing Josex "Gangsta way of doing LC", then fill new syringes with the LC (don't want to use the same LC jar multiple times over a longer period of time).

Also, would it be a bad idea to do Josex tek before transfers (or maybe after one transfer to make sure it's healthy)? I'm thinking that I want as many genes as possible so the chances that I get a fruit with traits I'm searching for will be higher.

I read about storing plates vertically around here. Apparently bacteria can't grow uphill so good :grin: plus the baccy juices will run down to the edge instead of over the top of your culture. Or that is my (mis)understanding of it.

Exactly that. I saw that Beef was storing plates on their side to help with condensation. I noticed recently with some baccy plates that the mush myc did grow upwards while the bacteria ran downwards.

I have also observed that pan myc that grow upward seems to be more robust, but that is more subtle and possibly just my own bias.



Not sure if someone else has written anything more up about it, though.

I'd take transfers from around 7 o'clock and 8 o'clock where it looks rhizo, take rice grain sized transfers, transfer to 3 or 4 plates.

Rhizo isn't always better than tomentose, but it's easier to tell it from most mold. Also looks cool.

Grow those plates out, then post the best looking one or two here.

When you get more experience, you won't be as likely to need 4 plates for each transfer, but if you have the plates, do it. Gives you more options, and gives you more to learn from.

I don't think the wood does anything for cubes, but it does seem like a convenience thing. It seems like it would be easier to work with since you can pull the stick out with sterile tweezers then grab a transfer.

I have slants with popsicle sticks in them but the only thing I have on them right now is wild turkey tail which I think wood is probably beneficial for. Make sure you hydrate/sanitize the sticks in boiling water before use.

Hey All,

I plan to put these three plates to grain do they look OK to you?

The third plate has a minor bluing to the top most section of mycelium poking up from the agar plug. I've found this happening on the highest fluffy areas over several plates and believe it to either be mould or stress. I thought it would be worth testing one to see if a jar contams, although based on the consistent position the bluing occurs and general health of the rest of the plates, I'm leaning slightly towards stress, I guess I'll find out shortly.
