nasopharyngeal swabs work well. I just tried them on these tamps. Picked up and deposited spores very efficiently. I do wish they weren't plastic/nylon, I prefer degradable materials.

I went a little overboard on germ plates. I wasn't expecting them all to take off but here we are. This agar thing is pretty fun even though it has exposed how incredibly impatient I am. Any suggestions on which plates I should focus on for transfers?

They all look great. Pick areas of nice clean uniform growth and the spots with the most dense rhizomorphs

I kind of think this looks neat and uniform, but should I be worried about the different growth between around 4 - 8?



edit. Too me it looks fine, just different. And I still havenโ€™t tried figuring out how can I get rid of condensation. Itโ€™s not so much after pour. Iโ€™m just guessing the temp fluctuates quite a bit because of the central heating here.

edit edit. Just noticed that Iโ€™ve also somehow poured the plate a bit thinner there, maybe thatโ€™s all it is.

Looking to start with agar. Well, in the mushroom context - I'm a lab technician and I've worked in a couple of microbiology labs during my career, and I've worked with petri dishes and agar etc, so I'm not a complete noob :laugh:

I had a spore syringe from a reputable seller/company (fwiw, they're based in a country where shrooms are legal to cultivate and possess for personal use, and they sell spores and mycelium for a lot of different edible mushroom species), and I used what was left of the spores (after PF tek cakes) to try if a liquid culture would work. Turns out it did, it's several weeks old now and I've kept it stored in the cold.

The PFtek cakes failed, but I still have that LC. FIgured I might as well try that on some agar next, before trying to get another print or syringe.

Another question for you guys. Attempting a clone here, second transfer. Growth is all over the place. Where would you take transfers?



First instinct would be the rhizo growth on top of the left one, right? But is it too ropey?

Hey for this India woodlover where exactly is best place to catch that rhizomorfic growth 7 to 9 oclock??

Like middle of growth towards edge of plate?
Or at ends with the rhizo tips?

Or both?

My advice is to not toss the actual clone plate. Tissue plates are usually 1) totally clean if you did them right 2) what youโ€™re after genetically, so thereโ€™s no need to transfer and 3) most likely in the cabinet to produce plate pins.

Stipe tissue is sterile. Iโ€™ve never had a clone contam unless I did something super wrong.

Keep them bad boys. Transfer when you want to expand.

Doh.. Makes perfect sense to be honest. I kind of presumed that, I guess I was just a bit worried that I'm messing something up. Also it was one finicky hollow stemmed pf-cake's shroom. More of a practice one than anything of real value, to be honest.

So what do you think is going on here? These are F1, T1 swabs from a giant mutant from my mutant strain The Thing, which was a mutated clone of my Husky, which is a standard cube.







This was the original mutant, and what plates looked like.





And those tubs usually came out a bit like this



These T1 plates, despite being from a spore swab, are creating mutant looking pins. Finally I have some of this on grains, so I should find out the outcome within a few weeks.

I was honestly expecting a revert back to a more standard cube after going back to spores, but perhaps this F1 is going to be a reliable "strain". Are there cases of mutant strains yielding reliable mutant offspring via spores? I know enigma doesn't spore at all.
