SSLI
(Super Sticky Liquid Inoculant)
(name and idea inspired by Semi-Solid Liquid Culture)

800 ml distilled water. 

0.2% agar, (1.6 grams agar.)

x7x+ plate is germination plate from Oct 19, 2024

PE is from Dec 19, 2022 (!) Both were in fridge.

Hamilton Beach Power Elite Blender (58148A).
I used the Dice / Chop / Salsa pulse setting. The rest are too powerful.

Before that I tried 2 different magnetic stirrers, neither were strong enough to shred an agar plate.

I tried two blenders, the second one (got at Salvation Army) worked. I have to hold the bottom (top) of the jar while blending. It only took a couple seconds to blend. I use pint jars. I PC the jars and the grinder top I made. I will update how it works. 


The smart thing to do with these old plates would be to have taken a transfer to a new plate, but I'm out of empty plates.

This is also my first try with oats. I used Bod's oat tek (very simple).

I kind of soaked every grain of oats with SSLI, and rolled the jars to get every bit. But it should be fine, that's supposed to be fine with SSLC. I've seen wetter jars on here without adding anything like this. haha.

Are they clear enough to make a guess whether the tomentose ones are clean? I have some fresh plates that I just did cabin sequestering, but I'm hoping to put the PESH and Nat to grain next.

:thumbup: the PESH looks great, you can try the NAT, but I'd feel bad about saying SEND IT, another xfer if able.

It might just be NAT doing what NAT does.... W(ever)TF it wants to.

It's the least fluffy of all the nat cultures I made. The rest of them reached the edge of the plate and grew up the sides. They also grew really thick. I have 0 knowledge to base it on, but I'm thinking that those cultures may have a higher chance of overlay, that or there is something nasty hiding in it.

I've lost many tubs, so if it fails it won't be a heartbreak, so don't feel bad if you suggest to send it and it goes bad.

I have some Nats in a cabin sequester, so the round after next will be grown from a transfer from those.

Since I started from a swab, I'm gambling on overlay genetics no matter what I do. Even if this plate gives me a lackluster flush, but doesn't overlay, it will advance my goals.

I'm just thrilled that I can take a halfway decent picture for once. I found a book light that I can adjust to light the plates from the bottom/side. Hoping to show off a decent culture once I have one.

The pics are good. 👍.

Even if the Nats overlay (and that might be all that's going on in those plates too), the overlay is still active and they're still worth growing.

Ugh. I’m convinced my plates are getting a lot of water in them because the lids aren’t flat, they have a lip so water can pool at the top. Has anyone come to that conclusion before? I just ordered some of Sages plates he told me to get on amazon.

Edit: oh plus I’ve been letting them cool in the pc a few days. So a pool of water just dripping in 😂 at least I’m learning before my new ttbvi plate gets here

Actually the first time I made them, I had no holes and negative pressure just sucked in as much as it could 😂 bro at least I’m learning ok!

Dip a swab and use it as you would any swab or use a loop to streak.

Probably just using a small inoculation loop that can fit right down into the vial. Or, pour some of the clumps into a shot glass or another bigger vial with more sterile water and mix it up. Sterilize the loop first, then swirl it around to pick up a tiny amount of spores, then streak the agar plate with it. Or use a disposable sterile swab.

Hm, I do have some sterilized cryovials at the ready so maybe I’ll transfer over to those first.

Would it be wreckless if me to think I could safely keep whatever spore/liquid around for a while? Or should this be a one and done?

Metal loops are re-usable & more eco friendly IMO. You can sector out the dish better when streaking too, to further improve the odds of germinating and isolating a culture away from any contams, like this:

I've heard you can just pour the spore solution onto some foil, and let it dry back out again in front of a hood or in a SAB. Then treat it like a dry print again. It would probably last a lot longer that way if you wanted to store it for a longer time.

The growth looks good around the edges (to my mostly untrained eye), but the yellow-ish looking bits around the center are tricky - could be weird lighting, or myc eating the dye in the agar, or it might be contams.

Kanthal A1 24ga wire
Smok RGC RBA (will screw directly into the battery, and made to preheat your coils with an adapter before putting in the tank).
In that picture is an "Aspire CF mod", which lets me use my 18650 batteries, and isn't too bulky like some mods. I call it my "Noc Saber".

You can use whatever smaller 510 batteries you want too, like this:

The middle bit is just agar with a different nutrient mix. It’s from a germination plate, and was fairly bulked up for some incredibly stubborn spores.
