4/9 of the plates have contaminated around the edges.

I will note that petris can be difficult to pick up. Is there a technique to this? When trying to pick up a closed petri dish, my gloves invariably get under the edge (of course they do, it's the only way to grasp the bottom under the cover) and this can't be good for sterility. I imagine that I introduce contamination under the lip of the cover and then airflow from the flow hood blows them into the media.

Somethings wrong with my plates for sure.

Still no growth on either 3 and I touched spit to them when it was mentioned like 4 days ago, opened in dirty dusty air for 30 seconds over a week ago, nothing. This is concerning to me, I've been having some confusing trouble with agar recently and this is the cherry on top lmao.

Bought the maltoferm 1001, gonna do a test batch to see if it hotbreaks but also going to try a 1.5% agar concentration unless anyone's got any suggestions

I have a suggestion, but I may get shit on for it. When I first started agar, absolutely nothing would grow on it. Not bacteria, not mold, not cube myc. Dead plates. Dropped my PC time to 20-30 minutes and my plates started working.

I tried tons of recipes with no success. Lowering cook time solved all my issues and any recipe works now.

I am well aware that the sugars in my agar scientifically speaking can’t be caramelizing, and I’m not making that claim, but the only variable change that made my plates work was cook time. So if you’re PCing more than half an hour I suggest dropping it and seeing what happens.

I know stipe recommends 45 min but cook time is dependent on volume being sterilized. I follow these guidelines from phytotech labs with success.  The total time includes ramp up + 15 minutes, so if the total min cook time were 40min and it took 10 min to reach 15psi, I interpret this to mean, you would only need to cook for an additional 15 min.  If the min time was 20 min (such as sterilizing multiple no pours with 25ml of medium in each), you’d essentially be done after hitting 15psi and cooking for an additional 5 min..  I have used this method with success.  If you’re feeling up to it, run a test on a few no pours or a small vol of pours and just let them sit for a week. 


Edit - stipe just chimed in :lol:  Our charts are similar but his doesn’t include the note on ramp up + 15.

Including the ramp up time is odd, because it varies depending on factors such as the size of the pressure cooker, the starting temperature, and the wattage of the heat source. None of which would seem to have anything to do with sterilization effectiveness. Is it not the only requirement that all the liquid has to be at 121Β°C for 15 minutes?

No. The volume is essential; 15 mins isn't sufficient for 75 ml let alone 500-1000ml, which are the standard volumes used for agar/LC.

15 mins may be sufficient for k cups/no pour containers but not for media bottles.
The charts have been developed by laboratories using biological indicators to verify sterilization cycle effectiveness.

The charts are dependable, reliable, and incontrovertible.

It seems like it could be relevant for some of the agar work home cultivators do but im not educated enough to understand the scope

What are your thoughts on their note?  If I’m between 250-1000ml, I have no issues at 25 min which includes the 10 min ramp up + 15 based on 40min for that range.

I just read the abstract but the article seems specific to bacterial cultivation.

I think we can get away with quite a bit. 25 mins + heat up should kill everything. My personal recommendation for 45 mins for everything under 1000ml is overkill but covers a multitude of sins including complete suspension of solids.

Folks have been successfully running agar for 15-20 mins under certain circumstances while others have issues. Without having to troubleshoot each individuals particular problem, just about every conceivable problem can be resolved with a single standardized answer:

Increase cycle time to 45 mins;
Use distilled h2o;
Vent pc for 15.


By following that simple guideline I can exclude the possibility of incomplete sterilization regardless of volume or elevation for most people. Fix issues with partially dissolved powder/sediment, clarity, water quality, etc.

The one size fits all approach, though takes longer to cycle, is a one stop fix for most common issues withput havimg to think too hard about the individual variables. Conservation of effort being the net return.
