Early on I’ll do two small transfers on the same plate instead of burning a second plate looking for clean myc.

Going forward each transfer gets a single plate. Once it gets to testing.

Mystery swab T2: maybe Melmak, maybe Natalensis.

All three MS cultures, probably go to grain next weekend for a round of tests.

I’ve got another Nat culture from a known source and it has produced some crazy variations in myc morphology. Most have not been this gently tomentose, some have been downright weird lookin. So we’ll see. I’d be hyped as fuck on some Melmak either way.

…all the typical methods. To some degree it’s inevitable in these cups (you can see the tiny micro-drops on the walls in the pics above). But I don’t let it bug me too much. The most effective prevention strategy I’ve found is to reduce the temperature differential as much as possible between the air inside the cup when pouring/sealing the lid, and the temp of the air outside the cup while the media sets. So I never pour until my media is at or below 125F (I like softer recipes so it doesn’t gel as soon), I prefer to pour in a warm setting when humidity is low, and allow sealed cups to cool slowly by stacking warm, and storing where they can stay warm for a few days until ready to use. Some will always have condensation on the lid. I just set those aside and use the clear ones first, a week passes and when I’m ready to do more work, the others have mostly cleared up.

Agar advice wanted please:

This is T0 of JMF. Spore Print to PDA agar. This one grew out pretty fast.



This is also a T0 taken at the same time with (Corn syrup/Potato flakes) agar. This one looks better to me, but also grew out slower.


This is a T1 (mislabeled as T2) that I took from the first T0 above. The agar is MEA.


I read somewhere a while ago it's good to switch up your agar nutrients because if you don't, myc can become lazy as to what it eats. But, as you can see in the T1, it's super whispy, while the parent T0 is not.

Should I bother taking another transfer from the T1, or just start from base again and do a new set of T1's from the T0's. If so, which T0 would you choose?

These have been transferred 3 times. Two different species. Both sclerotia. I am new to this so, are these samples considered isolates? Should I be seeing more brown spots since this is sclerotia? And if so, why am I not seeing some? Thanks guys!

They're isolates in one way but not another.

Looks like you've isolated mycelium you want away from contaminates you don't want.

As far as being genetic isolates though, I think they are a ways off yet.

Chasing down a single set of isolated genes the way youre probably thinking isnt really super beneficial.

Having healthy clean cultures is the primary goal. Looks like youre on the right track with that. I've usually noticed stones forming on plates when theyre a lil more mature. If youre looking to transfer stones then just give em time and grab one when it forms.

Where is the best online source for bulk (over 100 but less than 500) agar plates?  Looking for cost and consistently sterile plates.

What's a good "soft" LME agar recipe?  Should I do 8g agar, 8 grams LME, and 400ml H20? 

I had issues with contams in my plates and chunky agar and condensation and not super clear agar last time, so I'm stepping up my game this time.

What type of filter and where is a good online source for flow hood filters?  Is there a good TEK to describe how to make a hood?  I don't need step by step so much, but more of the concepts behind a good hood design and why.

Does this look like it could be mycelium and mold?  A lot of my pasty plates look like this.  It seems pretty uniform but the myc looks really tomentose and it makes think it could be mold.  Sorry for the shitty pics, unfortunately this is about as good as it gets for pasty plates. All of these pics are of the same plate.

IDK, looks pretty good to me but I don't know a lot. I've got some that look the same and I think they are doing pretty well but again, I don't have a lot of experience with this. I'll be interested in the answers you get.

I agree with this. If you're looking for better genetic isolation cloning is far superior to doing multiple agar transfers originating from a spore plate.
