You're not wrong, they're 4.5". It's sufficient for now, I can always make 2.0 after I use it more and understand what I need better.

The larger the hole, the less air that is moved (bellows) as you take your arms in and out of the box.

These are T1's of original ms plates.  I have no idea what I am actually growing. 

Some of the myc/growth seems to be defined and some looks almost like a smooth cloud.  There appears to be myc, but there is a fuzzy layer on top of the myc.  Is this mold/contam? 

The PE strains seem to have the most amount of fuzzn and height.  Can I take transfers from any of these or am I better placed going to water agar to see if I can get cleaner myc?

Looks good to me but I'm far from an expert here.

Yeah, I do have one: get a better swab donor :lol:
(Seriously I did nothing fancy: I just "Z" streak the plate with the Q-tip. All the credits go to the person who sent it to me. Clean AF Q-tip Tek bb!)

I know this is the agar thread, but if you have a really bacterial swab that you need to salvage, Josex puck poke tek has an answer for swabs that works really well.

First grow! Straight grain inoculation didn't make sense, since it seems easy to contaminate and I was going to be working with a spore print, so wanted least waste as possible.

2022-10-11: dropped spores into 4 LMEA dishes
2022-10-13: got anxious reading the forum thinking that I didn't "smear" the spores, so I dropped into one additional LMEA dish and scratched the media up to help
2022-10-17: got anxious again seeing the lack of any white mycelium, cooked up some dextrose+starch+yeast agar and poured and inoculated 3 more dishes. Up to 8 now

2022-10-20: actually there was some growth, just I didn't see it, since it was mostly translucent! I got it on good authority though, that it is fine and even transfers can be done on it, so I did some.





The wedges seem pretty small, but I had little growth to work with, it was difficult to see due to translucency, but I will see if it works out and if not - there are more plates :laugh:
