basic agar question
    #5240750 - 01/29/06 11:58 PM (18 years, 21 days ago)

ive heard that you can germinate spores on agar, then transfer the myc. to a substrate for colinzation. is this true? if so does that simply mean that germination and development of myc. occurs faster on agar then on grain substrate?

If I understand your question right..Yes. You can let the spores germinate on the agar in the petri dishes. Once this is finished, you can take a sterilized scalpel and cut a square away from the agar and transfer it to your substrate. This will in fact cut out the germination time. You don't have as many inoculation points though. You could make a liquid culture and it would work the same way. There are a few teks on the shroomery for this. Basically, you just mix up your solution. Karo, honey, etc...Sterilize it, let cool. Then you can inoculate it just like a regular jar. It is best to put a marble in with this solution so you can shake the jar and break up the mycelium easier. After the spores have germinated in the liquid culture, shake it up a bit. You can then use a sterilized syringe to pull the solution out of this jar and inoculate your substrate.

The reason to use Agar is to isolate mycelium that has good growth patterns.
