Here’s a T2 I’m really liking the look of. I think after another transfer it should be good.



Then I have a question: this is a T4 and it keeps growing out the same way every time I transfer it. Aggressive growth in one direction and slow growth in the other. I always take the most organized sample I can from the aggressive side, but lo and behold it grows out the same way. Should I keep transferring? It’s definitely a little tiny bit less chaotic each time, but a fully lopsided T4 seems weird to my inexperienced hand.

You should use a pc to sterilize your glass Petris the pressure and steam is what sterilizes and kills any heat resistant endo spores if I’m not mistaken.

Fingers crossed I start seeing plate pins this week from a couple germ plates. I believe I see knots forming on a few. Here's a B+ I took four transfers from, it didn't contam and kept growing clean:

Two in the front are RustyWhyte germ plates looks like knots starting to form too:



So looks like my first pins will be plins! I'll take biopsies and then possibly put the poked plins on agar too just to see the growth difference. I'm a believer in using a needle biopsy where I can. It has worked great for me.

So, after a whole night of research, I found out that my PC only cooks at 10PSI, and I was sterilizing with 15PSI times, which is probably the cause of the bacterial contamination.

Any suggestions on the time I would need to sterilize dishes under 10psi? I've only found grain sterilizing time (4 hours), but nothing on petri/agar.

Wondering if having them in a sealed plastic bag is preventing the PC from fully venting around your dishes leaving you with an incomplete sterilization. If you have any mushroom growing bags try using those but leave it unsealed. It will seal itself on cooldown. If not you might try one round with foil just to see if that’s the issue.

Others might do this successfully though so I’m just throwing out an idea. I wrap my pre-pours in foil but I reuse that foil until it’s torn and unusable.

I PC my glass petris before pouring. I don't trust anything else.

Yeah I tried reusing foil, but it always has some micro torns that can only be seen if you place some light behind, so I stopped with that.

I actually don't believe that the bag prevents much, since agar inside them is supposedly boiling again from the temperature, and they still get as much condensation as when I used the foil.

Ps. I got similar contamination when I used the foil too. Now that I remember I had to discard a whole petri batch once a couple of months ago!

It's probably more. According to the pressure chart someone published here for grains for example, a jar that took 90min to sterilize under 15psi, would take 240min to do so under 10psi.... Thats like a 2.7X increase.

So for the 20min, theoretically it would be somewhere around 60min. But I'm not sure if the agar would resist so much boiling without evaporating the water from it?

Decided to start playing with selection and isolation, this is my favorite plate so far.

The culture was cloned and further selected from a B+ MSS grow.

I've been picking plins for a few days now. I also have 3 grain jars waiting for inoculation.

Can I just cut some wedges off this plate and stick in the jars?


Or should I wait and use new growth from the clones?


Not sure i did this right, this clone keeps growing and is now touching the plate cover

those photos are awesome - macro lens ftw :super:
you could take wedges from that plate if youd like
if it is a germ plate i personally wouldnt, but if its t1 or t2+ im sure it would be fine
overgrown but looks clean to me
